摘  要:采用SephadexG-50微柱离心法结合HPLC法测定了半乳糖化胆固醇配体修饰去甲斑蝥素脂质体(Gal-NCTD-Lips)的包封率,并进行质量评价。当离心力为1000 r/min,离心时间为1 min,Gal-NCTD-Lips上样量为0.2 mL时,微柱离心法能有效分离脂质体和游离药物。在所建立的色谱条件下,去甲斑蝥素在0.03~0.18 mg/mL范围内线性关系良好(r=0.9994),高、中、低三种浓度的加样回收率分别为98.5%、98.9%、97.9%,包封率为51.06%±2.65%(n=5)。3种浓度的凝胶微柱的加样回收率分别为101.8%、99.25%、101.1%,RSD分别为1.69%、1.95%、1.40%,结果显示该方法准确、快速,能用于Gal-NCTD-Lips包封率的测定。A sephadex(G-50)microcolumn centrifugation combined with HPLC method was established to determine the entrapment efficiency of galactosylated cholesterol ligand modified liposomes containing norcantharidin(GalNCTD-Lips).The liposomes and free drug can be separated effectively by microcolumn centrifugation method when the centrifugal force was 1000 r/min,the centrifugal time was 15 min and the loading amount was 0.2 mL.Under the applied chromatographic condition,the linear relationship of norcantharidin was good in the range of 0.03~0.18 mg/mL(r=0.9994)and the average recoveries of high,middle and low concentrations were 98.5%,98.9%,97.9%,respectively,and the entrapment efficiency of Gal-NCTD-Lips was 51.06%±2.65%(n=5).The recoveries of three concentrations of sephadex(G-50)microcolumn were 101.8%,99.25%,101.1%with RSD=1.69%,1.95%,1.40%,respectively.These results showed that the method was accurate and simple,and reliable for the determination of the entrapment efficiency of Gal-NCTD-Lips.
