摘 要:目的:筛选白花香莲解毒颗粒抑制乙型肝炎病毒(hepatitis B virus,HBV)复制的mRNA表达谱,旨在发现及鉴定新的具有抗HBV活性的mRNA。方法:采用mRNA芯片及基因芯片检测HepG2细胞空白对照组、HBV1.3D侵染HepG2细胞模型组和白花香莲解毒颗粒干预组的mRNA表达谱,并通过生物信息学进行数据分析,筛选出靶mRNA。采用qRT-PCR方法对筛选出的mRNA进行验证。结果:mRNA芯片共筛选出与白花香莲解毒颗粒抑制HBV相关的183个发生显著变化的mRNA,其中在模型组表达升高,同时在白花香莲解毒颗粒干预组表达降低的有88个,在模型组表达降低,同时在白花香莲解毒颗粒干预组表达升高的有95个。qRT-PCR对IL8、EDN1、ENO2、PPARA以及NFIB共5个目的基因进行验证,结果与芯片一致。结论:白花香莲解毒颗粒对肝细胞内的mRNA表达具有干预作用,能通过调控mRNA发挥抑制HBV的作用。Objective:To screen the mRNA expression profile that Baihua Xianglian Jiedu Granules can inhibit the replication of hepatitis B virus(HBV)to find and identify new mRNA with anti HBV activity.Methods:The mRNA expression profiles of the blank control group of HepG2 cells,the model group of HepG2 cells infected by HBV 1.3D and the intervention group of Baihua Xianglian Jiedu Granules were detected by mRNA chip and gene chip,and the target mRNA was screened by bioinformatics data analysis.The qRT-PCR was used to verify the selected mRNA.Results:A total of 183 significantly changed mRNA related to the inhibition of HBV by Baihua Xianglian Jiedu Granules were screened out by mRNA chip,among which,the expression increased in the model group,and decreased in the intervention group of Baihua Xianglian Jiedu Granules,and decreased in the model group,and increased in the intervention group of Baihua Xianglian Jiedu Granules.qRT-PCR verified 5 target genes of IL8,EDN1,ENO2,PPARA and NFIB,and the results were consistent with the chip.Conclusion:Baihua Xianglian Jiedu Granule has an inter-vening effect on the expression of mRNA in hepatocytes,and can inhibit HBV by regulating mRNA.
