摘 要:目的:观察石菖蒲对颅脑损伤模型大鼠认知功能障碍、血清炎症因子及s100B、NSE蛋白水平影响。方法:选取SD雄性大鼠120只,随机分为空白组、模型组、吡拉西坦治疗组(阳性组)、石菖蒲高剂量组、石菖蒲低剂量组,每组24只。空白组除外,其余动物复制颅脑损伤模型,成模24 h后,阳性组给予吡拉西坦组灌胃[3.6 g·(kg·d)^-1)],供试药组给予石菖蒲高剂量、低剂量[高剂量2 g·(kg·d)^-1,低剂量1 g·(kg·d)^-1]灌胃,每天1次,连续给药14 d,于第1、7、14天动物分批进行水迷宫实验检测逃避潜伏期,腹主动脉取血检测血清肿瘤坏死因子(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)、IL^-10、s100β、神经元特异性烯醇化酶(neuronspecific enolase,NES)水平;各组动物取脑组织,检测脑组织含水量。结果:与模型组比较,石菖蒲各剂量组血清中TNF-α、IL-6、s100β及NSE蛋白水平均显著下降,差异均有统计学意义(P<0.05或P<0.01)。与模型组比较,石菖蒲各剂量组逃避潜伏期均显著下降,差异均有统计学意义(P<0.05)。结论:中药石菖蒲可有效降低颅脑损伤动物模型中血清炎症因子的异常表达,降低血清S100B、NSE蛋白水平,减轻脑组织水肿的病理损伤,提高模型动物认知障碍,有效改善颅脑损伤病理损伤。Objective:To observe the effects of Chinese herbal medicine Shichangpu(Acorus Tatarinowii)on cognitive dysfunction,serum inflammatory factors and s100β,NSE protein levels in rats with craniocerebral injury.Methods:One hundred and twenty SD male rats were selected and randomly divided into blank group,model group,piracetam treatment group(positive group),Shichangpu high-dose group,Shichangpu low-dose group,24 in each group.Except the blank group,the rest of the animals reproduced a model of craniocerebral injury.After the model was established for 24 hours,the positive group was given gavage to the piracetam group[3.6 g·(kg·d)^-1)],and the test group was given high dose Low-dose[high-dose 2 g·(kg·d)^-1,low-dose 1g·(kg·d)^-1]intragastrically,once a day,continuous administration for 14 d,animals divided on day 1,7,and 14 Water maze experiments were performed in batches to detect escape latency,and blood was taken from abdominal aorta to detect serum tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),IL^-10,s100β,neuron-specific enolase(NES)levels;brain tissue was taken from each group of animals,and brain water content was measured.Results:Compared with the model group,the levels of TNF-α,IL-6,S100β and NSE protein in the serum of each group of Chinese medicine Shichangpu significantly decreased,and the differences were statistically significant(P<0.05 or P<0.01).Compared with the model group,the escape latency of each group was significantly decreased,and the differences were statistically significant(P<0.05).Conclusion:Chinese medicine Shichangpu can effectively reduce the abnormal expression of serum inflammatory factors,reduce the levels of serum S100β and NSE protein,reduce the pathological damage of brain tissue edema,improve the cognitive impairment of model animals,and effectively improve the pathology of brain injury damage.
