摘 要:目的:观察沙苑子总黄酮对去势大鼠骨质疏松模型血清钙(Ca)、血清磷(P)、骨代谢及生物力学的影响。方法:60只雄性大鼠随机平均分为空白组、模型组、仙灵骨葆组(500 mg·kg^-1)、沙苑子总黄酮高剂量组(200 mg·kg^-1)、沙苑子总黄酮中剂量组(100 mg·kg^-1)、沙苑子总黄酮低剂量组(50 mg·kg^-1)。动物双侧卵巢切除术建立骨质疏松模型,3 d后给药,连续给药90 d。末次给药2 h后动物腹主动脉取血,检测血清中骨钙素(osteocalcin,OCN)、Ca、P、骨保护素(osteoprotegerin,OPG)、骨特异性碱性磷酸酶(bone-specific alkaline phosphatase,BALP)、抗酒石酸酸性磷酸酶(tartrate-resistant acid phosphatase,TPACP5b)、Ⅰ型胶原交联N-末端肽(N-telopeptide of typeⅠcollage,NTX)、Ⅰ型前胶原羧基末端肽(carboxy-terminal telopeptide of typeⅠ,CTX-Ⅰ)水平。取动物右侧大腿股骨,双能X射线检测股骨段骨密度值,取左侧大腿股骨,生物力学检测仪检测骨力学强度。结果:与模型组比较,沙苑子总黄酮可降低模型动物血清中OCN、P、BALP、TPACP5b、NTX、CTX-I水平,升高血清Ca、OPG水平,增加模型大鼠股骨中骨密度及骨力学强度,差异均有统计学意义(P<0.01或P<0.05)。结论:沙苑子总黄酮可有效调节去势大鼠骨质疏松模型血清钙、磷水平、骨代谢及生物力学,改善骨质疏松病情。Objective:To observe the effects of total flavonoids of Shayuanzi(Astragalus complanatus)on serum calcium and phosphorus,bone metabolism and biomechanical characteristics of osteoporosis models in ovariectomized rats.Methods:Sixty male rats were divided into blank group,model group,Xianling Gufu group(500 mg·kg^-1),Shayuanzi total flavonoid high-dose group(200 mg·kg^-1),and Shayuanzi total flavonoid medium-dose group(100 mg·kg^-1),Shayuanzi(Astragalus complanatus)total flavonoid low-dose group(50 mg·kg^-1).The osteoporosis model was established by bilateral oophorectomy in the animals.The osteoporosis model was administered 3 days later and continued for 90 days.Blood was taken from the abdominal aorta of the animal 2 hours after the last administration,and serum osteocalcin(OCN),calcium(Ca),phosphorus(P),osteoprotegerin(OPG),and bone-specific alkaline phosphatase were detected.(Bone-specific alkaline phosphatase(BALP),tartrate-resistant acid phosphatase(TPACP5b),N-telopeptide of typeⅠcollage(NTX),type I procollagen carboxyl terminus Carboxy-terminal telopeptide of type I(CTX-I)levels.The femoral bone of the right thigh of the animal was taken,and the bone density of the femoral segment was measured by dual-energy X-rays.The femoral bone of the left thigh was taken,and the bone mechanical strength was measured by a biomechanical detector.Results:Compared with the model group,the total flavonoids in Shayuanzi could reduce the levels of OCN,P,BALP,TPACP5b,NTX,and CTX-I in the serum of model animals,increase the levels of serum Ca and OPG,and increase bone density and bone in the femur of model rats.The differences in mechanical strength were statistically significant(P<0.01 or P<0.05).Conclusion:Total flavonoids of Shayuanzi(Astragalus complanatus)can effectively regulate serum calcium and phosphorus levels,bone metabolism and biomechanical characteristics of osteoporosis models in ovariectomized rats,and improve osteoporosis.
