摘 要:目的:研究新生儿瓜前退黄散对溶血性黄疸模型大鼠的治疗作用及机制,为临床使用该方提供实验依据。方法:采用以乙酰苯肼(Acetylphenylhydrazine,APH)灌胃诱导大鼠,复制溶血性黄疸模型大鼠,鼻饲予以瓜前退黄散,并观察给药后大鼠的一般行为学与生物学状态,检测血清中谷丙转氨酶(Alanine Aminotransferase,ALT)、谷草转氨酶(Aspartate Aminotransferase,AST)、总胆红素(Total Bilirubin,TBIL)、胆汁酸(Total Bile Acid,TBA)、直接胆红素(Direct Bilirubin,DBIL)和间接胆红素(Indirect Bilirubin,IBIL)含量。结果:模型组各指标均明显高于空白组(P<0.05),并以间接胆红素升高为主,表明大鼠溶血性黄疸模型造模成功;与模型组比较,瓜前退黄散高、中、低剂量组干预后的黄疸大鼠血清TBIL、DBIL、IBIL、AST、ALT、TBA含量均显著降低(P<0.05);且中药高剂量组的TBIL、DBIL、IBIL、ALT、AST、TBA与空白组相比,差异不具有统计学意义(P>0.05);中药中剂量组的ALT、AST与空白组相比,差异不具有统计学意义(P>0.05);中药低剂量组的AST与空白组相比,差异不具有统计学意义(P>0.05),表明瓜前退黄散可降低APH所致的血清肝功能损害,降低黄疸水平。各组间差异无统计学意义(P>0.05),说明实验中选择的3种剂量瓜前退黄散均能有效治疗溶血性黄疸。结论:新生儿瓜前退黄散对溶血性黄疸模型大鼠具有明显的治疗作用,其退黄机制可能为与改善血清中TBIL、DBIL、IBIL、AST、ALT、TBA的水平有关,减轻肝脏损伤,为临床使用该放治疗新生儿溶血性黄疸提供了科学依据。Objective:To investigate the effects of Guaqian Tuihuang San(瓜前退黄散)on model rats with hemolytic jaundice and to explore the mechanism of prescription compatibility.Methods:The rats were induced by APH intragastrically,and Model rats with hemolytic jaundice were copied,Guaqian Tuihuang San was nasogastric fed.After treatment,the general biological status in rats was observed,and the serum levels of ALT,AST,TBIL,TBA,DBIL and IBIL were detected.Results:All indexes in the model group were significantly higher than those in the normal group(P<0.05),and indirect bilirubin were more increased,indicating that the hemolytic jaundice model was successfully constructed in rats.Serum levels that TBIL,DBIL,IBIL,AST,ALT and TBA in the jaundice rats in the high,medium and low dose groups were more reduced than the model group(P<0.05).Meanwhile,there was no difference in TBIL,DBIL,IBIL,ALT,AST and TBA between the high-dose group and the model group(P>0.05).There was no difference in ALT,AST between the medium-dose group and the model group(P>0.05),and the serum levels of AST showed no significant difference between the low-dose group and model group(P>0.05).The results showed Guaqian Tuihuang San could reduce APH-induced serum liver function damage and jaundice level.There was no statistically significant difference among the groups(P>0.05),indicating that the three doses of Guaqian Tuihuang San could effectively treat hemolytic jaundice.Conclusion:Guaqian Tuihuang San was effective on rats with hemolytic jaundice,and the mechanism may be related with the serum levels of TBIL,DBIL,IBIL,AST,ALT and TBA,by reducing liver injury,providing a scientific basis in clinic.
