摘  要:目的:评价基于增感屏的辐射光成像技术的基本性能。方法:在体外和体内2种方式上将增感屏辐射光成像与切伦科夫光成像进行对比研究。体外研究中,采用Eppendorf管进行光谱特性、与核素成像的定量相关性、空间分辨力、灵敏度以及组织穿透性方面的对比。体内研究中,在小鼠模型上与切伦科夫光成像进行成像性能的对比,并进行兔子腘窝淋巴结增感屏辐射光成像。结果:增感屏辐射光成像的光信号可以被临床常用放射性核素激发,例如18-氟(18F)、32-磷(32P)和99m-锝(99mTc),其光信号强度与核素成像定量结果呈线性相关,可以获得比切伦科夫光成像高2个数量级的灵敏度以及更低的能量损失。在移植瘤小鼠与兔子腘窝淋巴结的显像中,组织靶器官都观察到了光信号。结论:增感屏辐射光成像在信号强度、灵敏度以及穿透性方面均优于切伦科夫光成像,展示出非常强的潜在临床应用价值。Objective To evaluate the performances of radioluminescence imaging technology based on a commercial intensifying screen(RLISI).Methods The performances of RLISI and Cerenkov luminescence imaging(CLI)were compared in vitro and in vivo.In vitro,Eppendorf tube was used to compare the spectral characteristics,quantitative correlation with nuclide imaging,spatial resolution,sensitivity and tissue permeability.In vivo,RLISI had its imaging performance contrasted with that of CLI for the mouse model,and was executed for rabbit popliteal lymph nodes.Results The optical signals of RLISI could be generated by clinical radioisotopes including 18F,32P and 99mTc and its intensity was linearly correlated to that of nuclear imaging.Compared with CLI,RLISI generated stronger signals with 2 orders of magnitude higher sensitivity and extremely low energy loss in tissue.Optical signals were observed in the tissue target organs in both the transplanted tumor mice and rabbit popliteal lymph nodes.Conclusion RLISI is superior to CLI in signal intensity,sensitivity and penetration,and has clear translational potential for imaging radioactive tracers in the clinical setting.
