摘 要:目的探讨替米沙坦和氯沙坦对胰岛素抵抗(IR)大鼠冠状动脉脂联素受体和IR的影响。方法将40只雄性SD大鼠随机分为对照组(Con组)、胰岛素抵抗组(IR组)、替米沙坦干预组[TEL组,5 mg/(kg·d)]和氯沙坦干预组[LOS组,10 mg/(kg·d)]。测定并比较各组大鼠空腹血糖、总胆固醇(TC)、三酰甘油(TG)和空腹胰岛素、血清脂联素(APN),并计算胰岛素敏感指数(ISI)。检测冠状动脉APN、脂联素受体1(AdipoR1)和脂联素受体2(AdipoR2)mRNA和蛋白的表达。结果与IR组比较,TEL组和LOS组血清APN水平均增加(P<0.05),且TEL组TG水平、空腹血糖和ISI低于IR组(P<0.05)。IR组冠状动脉APN mRNA、AdipoR1 mRNA、AdipoR2 mRNA表达低于Con组(P<0.05),TEL组和LOS组冠状动脉APN mRNA、AdipoR1 mRNA、AdipoR2 mRNA表达较IR组增高(P<0.05)。与IR组比较,TEL组和LOS组冠状动脉APN、AdipoR1和AdipoR2表达增加(P<0.05)。结论替米沙坦和氯沙坦均能增加IR大鼠血清APN水平,抑制冠状动脉APN、AdipoR1和AdipoR2表达的下降。Objective To explore the influence of telmisartan and losartan on adiponectin(APN)receptors(AdipoR)in rats with insulin resistance(IR).Methods Forty male sprague-dawley(SD)rats were randomly divided into control group(Con group),insulin resistance group(IR group),telmisartan intervention group[5 mg/(kg·d),TEL group]and losartan intervention group[10 mg/(kg·d),LOS group].Blood pressure(BP),APN,fasting blood glucose(FBG),fasting serum insulin(FSI),triglyceride(TG),total cholesterol(TC),and ISI were observed.The mRNA and protein expressions of AdipoR1,AdipoR2,and APN in rats coronary arteries(RCAs)were detected by reverse transcription-polymerase chain reaction(RT-PCR)and Western blot.Results Compared with Con group,the mRNA and protein expressions of AdipoR1,AdipoR2,and APN in IR group were significantly decreased(P<0.05).Compared with IR group,APN in TEL group and LOS group were significantly increased(P<0.05),the levels of TG and FBG in TEL group were decreased(P<0.05),ISI was improved(P<0.05).The expression of APN,AdipoR1,and AdipoR2 in TEL group and LOS group were increased(P<0.05).Conclusion Telmisartan and losartan could increase the levels of APN and inhibit the expressions of APN,AdipoR1,and AdipoR2 in the coronary artery of IR rats.
