摘 要:目的观察黄芪多糖对变应性鼻炎小鼠转录因子T-bet/GATA-3表达的影响,探讨黄芪多糖对其调节作用。方法30只Balb/c小鼠随机分成正常对照组、变应性鼻炎组和黄芪多糖组,通过OVA致敏建立变应性鼻炎动物模型,造模成功后,黄芪多糖组在第15 d给以黄芪多糖组200mg/kg灌胃,共2周。观测各组小鼠症状表现,HE染色检测鼻腔黏膜中嗜酸性粒细胞总数;ELISA检测小鼠血清中s Ig E、IL-4、IL-5、IFN-γ含量;Western blot法检测鼻黏膜组织中T-bet、GATA-3蛋白的表达。结果小鼠经黄芪多糖干预后,变应性鼻炎症状缓解,黄芪多糖可以降低鼻黏膜嗜酸性粒细胞数量,血清中s Ig E、IL-4、IL-5水平下降(P<0.01),IFN-γ水平上升(P<0.05)。与对照组、黄芪多糖组比较,AR组中T-bet蛋白含量明显减低,而GATA-3明显升高(P<0.01)。结论转录因子T-bet/GATA-3在变应性鼻炎中表达失衡,黄芪多糖可以抑制变应性鼻炎的鼻腔黏膜嗜酸性粒细胞炎症,纠正T-bet/GATA-3平衡,从而起治疗作用。Objective To investigate imbalanced expression of transcription factors T-bet and GATA-3 and the effects of Astragalus Polysaccharide on them in rat allergic rhinitis models.Methods Thirty Balb/c mouse were randomly divided into control group,allergic rhinitis group(Group AR),and Astragalus Polysaccharide group(Group ASP).The allergic rhinitis model was induced with OVA.Astragalus polysaccharides group was given 200 mg/kg in Astragalus polysaccharide group at 15 d for 2 weeks.The total eosinophils in nasal mucosa were detected by HE staining,The concentrations of sIg-E,IL-4,IL-5,IFN-γin serum were measured by ELISA,and the expressions of T-bet and GATA-3 in the nasal mucosa were detected by Western blot.Results After intervention with Astragalus polysaccharide,the symptoms of allergic rhinitis in mice were alleviated.Astragalus polysaccharide could reduce the number of eosinophils in nasal mucosa,decrease the levels of sIgE,IL-4 and IL-5 in serum(P<0.01),and increase the level of IFN-gamma(P<0.05).Compared with the control group and astragalus polysaccharide group,the content of T-bet protein in AR group decreased significantly,while GATA-3 increased significantly(P<0.01).Conclusions The expressions of transcription factors T-bet and GATA-3 are imbalanced,APS can inhibit the airway inflammation and correct the T-bet/GATA-3 balance in allergic rhinitis.
