摘  要:蒙古冰草(Agropyron mongolicum Keng)是禾本科冰草属多年生草本植物,对干旱、寒冷及风沙具有很强的抵抗力,是干旱草原和荒漠地带的重要牧草。iTRAQ(isobaric tags for relative and absolute quantification)技术即同位素相对标记与绝对定量技术,是近年来最新开发的一种新的蛋白质组学定量研究技术,具有分离能力强、定量结果可靠、灵敏度高、可检测出低丰度蛋白等特点。以蒙古冰草为材料进行干旱处理12d后复水处理,分别在处理0,4,8,12,16d取样,利用iTRAQ技术分析蒙古冰草叶片在干旱胁迫及复水处理下蛋白质组学的变化,并进行生物信息学分析,研究结果表明:在干旱情况下持续上调的蛋白15个,持续下调的蛋白6个,在干旱复水处理中发现有1个基因(Q40001)是在干旱处理中处于下调状态,但是在复水处理后处于上调状态,通过查询蛋白的注释信息发现,该蛋白是一种镁离子螯合酶,其含量的变化与叶绿素的变化趋势相符。Agropyron mongolicum Keng is a kind of perenial herb in Gramineae, It is an important pasture in arid grasslands and desert areas for its strong resistance to drought, cold and wind. iTRAQ, also known as isobaric tags for relative and absolute quantification, which is a newly developed technique for quantitative study in proteomics, has an advantage of strong separation ability, reliable in result, high sensitivity and can detect low abundance protein. In this experiment, The Agropyron mongolicum Keng was disposed in drought for 4, 8 and 12 days and rehydration for 4 days after 12 days drought treatment, and then their leaves proteomics variation and bioinformatics in drought and rehydration was analyzed by iTRAQ technique. The result revealed that 15 up-regulated proteins and 6 down-regulated proteins in dry condition,one gene from the rehydration sample of Agropyron called Q40001 was decrease in down-regulated status in drought, while it was up-regulated status after rehydration. It is confirmed that this protein is a kind of Magnesium ion chelase, which variation is corresponded with the variation of chlorophyll content.
