摘 要:目的观察血府逐瘀汤对血管紧张素Ⅱ(AngⅡ)诱导大鼠骨髓源内皮祖细胞(EPCs)衰老、迁移功能及miR-34a表达的影响。方法体外培养EPCs,加入100 nmol/L AngⅡ诱导细胞衰老,建立内皮祖细胞衰老模型,随机分为正常组、模型组、miR-34a抑制剂组、5%血府逐瘀汤含药血清组(5%含药血清组)、10%血府逐瘀汤含药血清组(10%含药血清组)、15%血府逐瘀汤含药血清组(15%含药血清组)。β-半乳糖苷酶染色法检测内皮祖细胞衰老;Transwell小室检测EPCs迁移功能;qRT-PCR法检测miR-34的表达。结果与正常组比较,模型组EPCs衰老数量明显增多(P<0.01),EPCs的迁移数量减少(P<0.01),miR-34a表达量明显升高(P<0.01);与模型组比较,miR-34a抑制剂组和5%含药血清组、10%含药血清组、15%含药血清组EPCs衰老数量减少(P<0.01),EPCs的迁移数量明显增多(P<0.01),miR-34a抑制剂组和10%含药血清组、15%含药血清组中miR-34a的表达量降低(P<0.01);与miR-34a抑制剂组比较,5%含药血清组、10%含药血清组、15%含药血清组EPCs衰老数量增多(P<0.01),EPCs的迁移数量明显减少(P<0.01),miR-34a表达量明显升高(P<0.01);与5%含药血清组比较,10%含药血清组、15%含药血清组EPCs衰老数量明显减少(P<0.01),EPCs的迁移数量明显增多(P<0.01),miR-34a表达量明显降低(P<0.01);与10%含药血清组比较,15%含药血清组EPCs衰老数量明显减少(P<0.01),EPCs的迁移数量显著增多(P<0.01),miR-34a表达差异有统计学意义(P<0.01),3个含药浓度组比较,在不同干预时间点比较差异无统计学意义(P>0.05)。结论血府逐瘀汤能够延缓大鼠EPCs的衰老,促进EPCs迁移,下调miR-34a的表达。Objective To observe the effects Xuefu Zhuyu decoction(XZD)on angiotensinⅡ(AngⅡ)-induced senescence,migration function and miR-34a expression of bone marrow-derived endothelial progenitor cells(EPCs)in rat.Methods EPCs were cultured in vitro,and 100 nmol/L AngⅡwas used to induce cell senescence.The model of EPCs senescence was established.EPCs was divided into normal group,model group,miR-34a inhibitor group,5%XZD serum group,10%XZD serum group,and 15%XZD serum group.β-galactosidase staining was used to detect the senescence of endothelial progenitor cells.The migration ability of EPCs was detected by Transwell chamber.The expression of miR-34 was detected by qRT-PCR.Results Compared with the normal group,the number of aging EPCs in model group increased significantly(P<0.01).The number of EPCs migrated in the model group was significantly decreased(P<0.01),while the expression of miR-34a was significantly increased(P<0.01).Compared with the model group,the number of aging EPCs in the miR-34a inhibitor group and 5%,10%,and 15%XZD-containing serum group were significantly decreased(P<0.01),while the number of EPCs migrated was significantly increased(P<0.01),the expression of miR-34a in the miR-34a inhibitor group and 10%and 15%serum containing sera was significantly decreased(P<0.01).Compared with the miR-34a inhibitor group,the number of aging EPCs in 5%,10%and 15%serum containing sera were significantly increased(P<0.01).The number of migrated EPCs was significantly decreased(P<0.01),while the expression of miR-34a was significantly increased(P<0.01).Compared with the 5%XZD serum group,the number of aging EPCs in 10%and 15%XZD serum containing sera were significantly decreased(P<0.01).The number of EPCs migrated was significantly increased(P<0.01),while the expression of miR-34a was significantly decreased(P<0.01).Compared with the 10%XZD serum group,the number of aging EPCs in 15%XZD serum containing sera was significantly decreased(P<0.01).The number of EPCs migrated was significantly increased(P<0.01)
