摘 要:目的:探讨四物汤对环磷酰胺和乙酰苯肼致血虚证模型小鼠骨髓细胞凋亡的影响及机制。方法:实验动物随机分为正常对照组、模型对照组、四物汤组和阳性药对照组。除正常对照组外,其余三组小鼠分别于给药后第2天、第5天皮下注射乙酰苯肼每日20 mg/kg、40 mg/kg,从第5天起,每日腹腔注射环磷酰胺40 mg/kg,连续4 d,制备血虚证小鼠模型。利用血细胞计数仪、流式细胞术和蛋白免疫印迹技术分别检测各组小鼠外周血象、骨髓细胞凋亡率及骨髓细胞Bcl-2蛋白表达。结果:与模型对照组相比,四物汤组血红细胞数升高非常显著(P<0.01),白细胞数和血红蛋白含量均无显著变化(P> 0.05);四物汤组小鼠骨髓细胞凋亡率下降非常显著(P<0.01),Bcl-2蛋白表达量显著增加(P<0.05)。结论:四物汤能够促进小鼠骨髓细胞中Bcl-2蛋白的表达,抑制骨髓细胞凋亡,提高外周血象,此可能是其发挥补血作用的机制之一。Objective:To investigate the effect and mechanism of Siwu Decoction (四物汤) on mice with blood deficiency induced by cyclophosphamide and acetylphenylhydrazine. Methods:The experimental animals were randomly divided into the normal control group,the model control group,the Siwu Decoction group and the positive drug control group. Except the normal control group,mice in each group were injected subcutaneously with 20 mg·kg-1·d-1 and 40 mg·kg-1·d-1 of acetylphenylhydrazine on the second day and the fifth day after administration respectively. Starting from the fifth day,40 mg·kg-1·d-1 of cyclophosphamide was injected intraperitoneally for four consecutive days,then the mice model of blood deficiency syndrome was prepared. Peripheral blood,apoptosis rate of bone marrow cells and Bcl-2 protein expression of bone marrow cells in each group were detected by hemocytometer,flow cytometry and Western blotting. Results:Compared with the model control group,the number of red blood cells of mice in the Siwu Decoction group increased significantly(P<0.01),and the number of white blood cells and hemoglobin content did not change significantly(P> 0.05). The apoptosis rate of bone marrow cells in the Siwu Decoction group was significantly decreased(P<0.01),and the expression of Bcl-2 protein was significantly increased(P<0.05). Conclusions:Siwu decoction can promote the expression of Bcl-2 protein in bone marrow cells of mice,inhibit the apoptosis of bone marrow cells and improve the peripheral hemogram,which may be one of the mechanisms of its effect on tonifying blood.
