一贯煎对大鼠肝星状细胞雌激素受体的影响

摘　　要：目的:观察一贯煎含药血清对大鼠肝星状细胞(HSC-T6)雌激素受体α(ERα)、雌激素受体β(ERβ)2种亚型基因表达的影响,探讨其抗肝纤维化的作用机制。方法:体外培养活化的大鼠HSC-T6细胞,分为空白组、雌二醇组和一贯煎高、中、低剂量组,共5组,空白组加入空白鼠血清,药物组加入相应的含药血清进行干预,培养24 h、48 h、72 h后,分别收集细胞和上清液,保存备用。MTT法检测细胞增殖情况;ELISA法检测细胞上清液中Ⅰ型胶原和Ⅲ型胶原含量;Real time-PCR方法检测ERα、ERβmRNA的表达。结果:一贯煎高剂量组表现对细胞增殖有明显的抑制作用,能够降低细胞上清液中Ⅰ型胶原和Ⅲ型胶原含量,且作用较雌二醇显著(P<0.05);一贯煎在提高HSC-T6细胞ERα、ERβmRNA的表达方面作用较雌激素含药血清组弱,差异无统计学意义。结论:一贯煎能够抑制肝星状细胞活化,其机制可能是通过提高肝星状细胞的雌激素受体表达而起作用。Objective:To investigate the effects of Yiguan Jian on ERαand ERβof hepatic stell cells(HSC-T6)in rats,and to explore its action mechanism against hepatic fibrosis.Methods:In vitro culture of rat hepatic stellate cell line HSC-T6,the cells were divided into the blank control group,E2 group and the groups of Yiguan Jian(low-dose,medium-dose and high-dose).The MTT method was used to detect the inhibition of cell viability when cells were treated with different doses of serum containing Yiguan Jian for 24 h,48 h and 72 h respectively.Contents of collagenⅠand collagenⅢwere detected by ELISA method.Levels of ERαmRNA and ERβmRNA were measured by Real-Time PCR.Results:Compared to the blank control group,the high-dose Yiguan Jian and E2 inhibited the proliferation of HSC-T6 and down-regulated the expression of collagenⅠand collagenⅢsignificantly(P<0.05);the effect of high-dose Yiguan Jian was more significant than E2(P<0.05).The expressions of ERαmRNA and ERβmRNA were increased in the high-dose group of Yiguan Jian and in E2 group than those in the blank control group(P<0.05);there was no statistical difference between high-dose group of Yiguan Jian and E2 group(P>0.05).Conclusion:Yiguan Jian drug-containing serum can effectively inhibit the proliferation of HSC-T6,and alleviate hepatic fibrosis,the mechanism may be partially related to regulation of ERαmRNA and ERβmRNA.
