摘  要:目的:观察电针干预对尾部悬吊小鼠比目鱼肌Foxo3a蛋白及mRNA表达的影响。方法:将28只小鼠随机分成4组,即正常组、尾吊组、提前电针组和电针组,每组7只。正常组全程常规饲养;尾吊组前14 d常规饲养,后14 d尾部悬吊造模;提前电针组为在尾吊组处理基础上于前14 d加用每天电针;电针组为在尾吊组基础上于后14 d加入每天电针干预。电针每日1次,每次15 min。28 d后分别取两侧比目鱼肌,采用蛋白质印迹(Western blot)法检测小鼠比目鱼肌Foxo3a蛋白的表达,实时荧光定量聚合酶链式反应(Real-time PCR)法检测Foxo3a mRNA的表达。结果:尾吊组与正常组比,Foxo3a的Western blot蛋白表达和mRNA的表达升高(P<0.05);电针组与尾吊组比Foxo3a的Western blot蛋白和mRNA表达下降(P<0.05),提前电针组与尾吊组比Foxo3a的Western blot蛋白和mRNA表达亦下降(P<0.05),但两组仍较正常组高(P<0.05);Foxo3a蛋白和mRNA表达在电针组和提前电针组间比较没有统计学意义。结论:电针对可能参与骨骼肌萎缩的Foxo3a有一定干预作用。Objective:To observe the effect of electro-acupuncture(EA)on the protein and mRNA expressions of Foxo3a of soleus in tail-suspended mice.Methods:The mice were randomly divided into the blank group,the model group,the pre-intervention group(EA 14 days before modeling)and the EA group(EA 14 days after modeling),with 7 mice in each group.The weightlessness model was simulated by tail suspended in mice.EA(1 mA,20~30 Hz)was applied to bilateral Zusanli(ST36)and Yanglingquan(GB34)for 15 min.After 28 days,Western blot and Realtime PCR were used to detect the protein and mRNA expressions of Foxo3a in soleus of mice.Results:The protein and mRNA expressions of Foxo3a were increased obviously in the model group compared to those in the blank group(P<0.05).The expressions were significantly decreased after electro-acupuncture intervention and pre-intervention of acupuncutre(P<0.05);however the expressions were still higher in the two groups than those in the blank group(P<0.05).There were no statistical differences in the expressions between the two intervention groups(P>0.05).Conclusion:EA has a certain intervention effect on Foxo3a expression that may be involved in simulated weightless skeletal amyotrophy.
