摘 要:目的探索神经妥乐平(Neurotropin,NTP)对H2O2(Hydrogen peroxide)诱导的C17.2神经干细胞凋亡的保护作用及其可能的机制。方法用CCK8法检测细胞活力,荧光显微镜观察细胞内活性氧的产生,并用qPCR检测p53、Bax、bcl-2的mRNA表达。结果C17.2神经干细胞的活力随H2O2的浓度增加而下降。用1000μM H2O2处理干细胞2 h后细胞的活力明显降低,细胞内活性氧生成增加,p53、Bax的mRNA表达升高,bcl-2的mRNA表达下降,上述结果与对照组相比,差异均有统计学意义(均P<0.05)。而用神经妥乐平(NTP)预处理24 h则可以明显改善由H2O2诱导的细胞凋亡损伤,提高细胞活力,细胞内活性氧水平减低,并且抑制p53、Bax的mRNA表达,上调bcl-2的mRNA表达,上述指标与对照组相比,差异均有统计学意义(均P<0.05)。结论H2O2能诱导C17.2神经干细胞凋亡,而神经妥乐平(NTP)能改善这种损伤,该作用可能与其抑制p53、Bax基因表达、促进bcl-2基因表达有关。Objective To investigate the protective effect and its possible mechanisms of Neurotropin(NTP)on hydrogen peroxide(H2O2)-induced apoptosis in C17.2 neural stem cells.Methods Cell counting kit-8(CCK-8)assay was used to test the cell viability of each group.The intracellular ROS levels of cells were observed under a fluorescence microscope.The mRNA levels of p53,Bax,Bcl-2 were detected by qPCR.Results The vitality of C17.2 neural stem cells decreased with the increase of the concentration of H2O2.After treated with 1000μM H2O2 for 2 h,the viability of the cells significantly decreased,the production of reactive oxygen species increased,the mRNA expression of p53 and Bax increased,and the mRNA expression of Bcl-2 decreased.The above results were statistically significantly different compared with the control group(P<0.05).It can significantly improve the apoptosis induced by H2O2 to pretreat with Neurotropin(NTP)for 24 h,increase cell viability,decrease the levels of intracellular reactive oxygen species,and inhibit the mRNA expression of p53 and Bax,and up-regulate the mRNA expression of Bcl-2,the above indicators were statistically significantly different compared with the control group(P<0.05).Conclusion Neurotropin demonstrated a potential neuroprotective effect on apoptosis-induced by hydrogen peroxide in C17.2 neural stem cells,which might be related to p53,Bax,and Bcl-2.
