摘 要:将楸叶泡桐四倍体的叶片和叶柄分别置于含有不同质量浓度的6-BA(6~20 mg/L)和NAA(0.1~1.1 mg/L)的组合培养基上诱导再生植株,筛选最佳外植体与最适培养基,建立同源四倍体楸叶泡桐体外植株再生系统。结果表明,同源四倍体楸叶泡桐体外植株再生的最佳外植体是叶片;其幼苗叶片愈伤组织诱导的最适培养基为MS+14 mg/L 6-BA+0.3 mg/L NAA;愈伤组织芽诱导的最适培养基为MS+16 mg/L 6-BA+0.3 mg/L NAA;幼芽生根的最适培养基为1/2MS。The system of in vitro plantlet regeneration from the leaves of autotetraploid of Paulownia catalpifolia was established in order to lay foundation for the breeding of new variant of Paulownia plants.The induction of in vitro plantlet regeneration from the leaves and peioles of autotetraploid with different hormones and concentrations(0.1-1.1 mg/L NAA,6-20 mg/L 6-BA)of media were investigated in order to screen the optimal explants and media.The result indicated that the leaves were the optimal explants for in vitro plantlet regeneration of autotetraploid Paulownia catalpifolia plant.Moreover,the optimal media of callus induction from Paulownia catalpifolia was MS+0.3 mg/L NAA+14 mg/L 6-BA,and that MS+0.3 mg/L NAA+16 mg/L 6-BA might be optimal its shoot induction media;and the optimal rooting media of the shoot was 1/2MS media.
