摘  要:为获得大量栎叶绣球的无菌苗,本研究以栎叶绣球‘雪花’(Hydrangea quercifolia’Snow Flake’)的叶片为试验材料,通过不同浓度的激素组合处理。结果表明,最适宜的愈伤组织诱导培养基为:MS+1.5 mg/L6-BA+0.2 mg/L NAA,出愈率约为80.7%;最适宜的不定芽分化培养基为:MS+2 mg/L 6-BA+0.1 mg/L IBA,诱导率约为90.3%;最佳的增殖培养基为:MS+2.5 mg/L 6-BA+0.1 mg/L IBA,增殖率约为5.2;最佳的壮苗培养基为:MS+0.5 mg/L 6-BA+0.15/0.2 mg/L IBA,幼苗平均高度约为4.57 cm;最佳的生根培养基为:1/2 MS+0.4 mg/L IBA,平均生根率约为88.3%,平均根长约为6.5 cm。本研究初步建立了栎叶绣球‘雪花’的再生体系,为其扩大繁殖和之后的遗传转化研究提供了帮助。To obtain a large number of aspetic seedlings of Hydrangea quercifolia, we used the leaves of Hydrangea quercifolia ’Snow Flake’ as test materials, treated by different concentrations of hormones. The results showed that the optimum callus induction medium was: MS+1.5 mg/L 6-BA+0.2 mg/L NAA, and the induction rate was about 80.7%. The optimum adventitious shoot differentiation medium was: MS+2 mg/L 6-BA+0.1 mg/L IBA, the induction rate was about 90.3%. The best proliferation medium was: MS+2.5 mg/L 6-BA+ 0.1 mg/L IBA,the proliferation rate was 5.2. The best medium for seedling cultivation was: MS+0.5 mg/L 6-BA+0.15/0.2 mg/L IBA, and the average height of the plant was about 4.57 cm. The best rooting medium was: 1/2 MS+0.4 mg/L IBA,average rooting rate was about 88.3% and the average root length was about 6.5 cm. This study initially established the regeneration system of the Hydrangea quercifolia ’Snow Flake’, which is helpful to the study of expanded reproduction and subsequent genetic transformation.
