摘  要:徒手切片常用于植物细胞结构的研究,但传统徒手切片方法对操作技术要求较高,且需要经历复杂的脱色和染色.利用双面剃须刀将新鲜水稻叶片切成薄片,分别在可见光和紫外光下观察显微结构.结果显示,叶片横切面在蓝色荧光下,可以获得清晰的表皮细胞、维管束、泡状细胞大小和数目的图像.该图像满足计量分析.采用改进的徒手切片方法操作简便、快速,适合水稻叶片维管束和泡状细胞变异的快速筛选.Free-hand sectioning is often used in the study of plant cell structure.However,the traditional method of free-hand sectioning needs high technical requirements and complicated destaining and staining.The free-hand sectioning to make slices of rice leaves was improved using two pieces of shaver,and the cell structure was observed through visible and ultraviolet light microscope.The result showed that the clearest cell structure and vascular bundle of leaves were obtained under blue fluorescence,which is fit to analysis the number and size of bulliform cell and cell in vascular bundle.Our results indicated that fluorescence microscopic observation for free-hand section was a very simple and so-easy learned technique.
