ICP-AES法测定自闭症儿童头发和指甲中Pb等6种元素含量及其相关性分析

摘　　要：目的观察自闭症儿童与正常对照儿童头发和指甲中金属含量,探讨其与自闭症病因相关性。方法采用电感耦合等离子体原子发射光谱仪对样本中金属离子浓度进行测定。实验结果采用t检验进行统计分析。结果自闭症儿童组指甲中铅(Pb)含量为11.51±5.97μg/g,对照组为2.92±1.82μg/g,病例组指甲中Pb含量远高于对照组(P<0.01),自闭症儿童组头发中铅(Pb)含量为4.62±1.11μg/g,对照组为1.50±0.69μg/g,病例组头发(指甲改为头发)中Pb含量远高于对照组(P<0.01)。自闭症儿童组指甲中铜(Cu)含量为22.19±3.94μg/g,对照组为6.59±1.14μg/g,病例组指甲中Cu含量远高于对照组(P<0.01)。自闭症儿童组指甲中锌(Zn)含量为118.52±28.93μg/g,对照组为157.29±7.32μg/g,病例组指甲中Zn含量低于对照组(P<0.01)。自闭症儿童组指甲中铁(Fe)含量为160.97±13.75μg/g,对照组为177.81±16.22μg/g,病例组指甲中Fe含量低于对照组(P<0.05)。自闭症儿童组头发中除了铅元素之外,其他各种金属含量与对照组相比,未发现统计学差异(P>0.05)。结论自闭症患者头发和指甲中Pb含量升高,指甲中Zn/Cu比值降低,Fe含量降低,Ca和Mn的含量在头发和指甲中均没有明显差异,为探索自闭症的病因及诊断提供新思路。Objective This study aimed to assess the metal content in hair and nails of autistic children and healthy children,and to explore its correlation with the etiology of autism.Methods The concentration of metal ions in the samples was determined by inductively coupled plasma-optical emission spectrometer(ICP-OES).The experimental results were statistically analyzed by t test.Results Relative expression of Pb in cases and controls in nails was 11.51±5.97μg/g and 2.92±1.82μg/g respectively,in hair was 4.62±1.11μg/g and 1.50±0.69μg/g respectively,with a significant difference between them(P<0.01).Relative expression of Cu in cases and controls in nails was 22.19±13.94μg/g and 6.59±0.14μg/g respectively,with a significant difference between them(P<0.01).The Zn content in the nails of children with autism was 118.52±28.93,and that in the control group was 157.29±7.32.The Zn content in the nails of the patients was lower than that in the control group(P<0.01).The Fe content in the nails of children with autism was 160.97±13.75,and that in the control group was 177.81±16.22.The Fe content in the nails of the patients was lower than that in the control group(P<0.05).But there was no statistical difference in the content of other metals in the hair of in cases and controls(P>0.05).Conclusion The content of Pb and Cu in nails of autistic patients increases,and the content of Zn and Fe in nails decrease,which provides new ideas for exploring the etiology and diagnosis of autism.
