摘 要:[目的]鉴定铁皮石斛与金钗石斛杂交F1代和回交后代的真假性。[方法]以14株铁皮石斛与金钗石斛杂交F1后代及18株以铁皮石斛为母本回交后代为试验材料,从24对特异性SSR引物中筛选引物用以鉴定。[结果]从24对SSR基因组引物中,筛选出4对双亲多态性引物。用该4对石斛特异性引物可从14株杂交F1群体中鉴定出11株真杂交种,占杂交群体的78.6%;18株回交群体中,被4对引物同时鉴定出真杂种率达38.9%。[结论]利用SSR分子标记技术鉴定石斛种间杂交与回交是可行的,对提高石斛育种效率、缩短育种年限和石斛种质资源的保护与利用具有重要意义。[Purpose] To verify the authenticity of true F1 and backcross progenies of Dendrobium catenatum×D.nobile. [Method] Fourteen putative F1 hybrids of D.catenatum×D.nobile and 18 backcross progenies with D.catenatum as the female parent were used as test materials. Twenty-four pairs of specific SSR primers were selected for identification. [Result] Out of the 24 Dendrobium specific SSR primer pairs, 4 primer pairs were found to be polymorphic to both parents. Using these four used to identify 11 F1 hybrids from the 14 putative hybrids F1 populations, accounting for 78.6% of the hybrid population;at the same time, a true backcross progeny rate of 38.9% was identified among the 18 putative backcross populations. [Conclusion] The SSR molecular marker technology was used to identify the true interspecific hybrids and backcross progenies, which is of great significance for improving the efficiency of Dendrobium breeding, shortening the breeding period, and protecting and utilizing the Dendrobium germplasm resources.
