摘 要:目的:探讨胃复方含药血浆对人胃癌BGC-823细胞的抑制作用及其机制。方法:MTT法检测细胞增殖抑制率,流式细胞技术检测细胞周期,RT-PCR及Western blot法检测细胞中垂体瘤转化基因(pituitary tumor transforming gene,PTTG)、骨髓细胞瘤病毒癌基因(cellular-myelocytomatosis viral oncogene, c-Myc)、人端粒酶逆转录酶(human telomerase reverse transcriptase, hTERT)的mRNA及蛋白表达水平。结果:胃复方含药血浆低、中、高剂量组的吸光度(OD)值依次降低并与作用时间负相关,抑制率(IR)依次升高并与作用时间正相关;胃复方含药血浆不同浓度组干预胃癌细胞72 h后,其G0/G1期细胞比率显著增加,并与其浓度正相关;S期和G2/M期细胞比率均显著减少,并与其浓度负相关。各组含药血浆作用于胃癌细胞72 h后,与空白血浆组比较,5-Fu组、中药组均对PTTG、c-Myc、hTERT的mRNA相对表达量、蛋白表达量有下调作用并呈剂量依赖性。结论:胃复方含药血浆可能下调PTTG、c-Myc、hTERT的表达以阻滞细胞周期进程,从而抑制人胃癌BGC-823细胞增殖达到抗肿瘤作用。Objective:To investigate the inhibitory effect and mechanism of gastric compound containing plasma on human gastric cancer BGC-823 cells. Methods: The inhibitory rates of gastric cancer cells on proliferation were measured by MTT;Flow cytometry was used to measure the cell cycle, The expression of pituitary tumor transforming gene(PTTG),cellular-myelocytomatosis viral oncogene(c-Myc)and human telomerase reverse transcriptase(hTERT) mRNA and protein in gastric cancer cells was detected by RT-PCR and Western bolt methods. Results: The optical density(OD) decreased in turn and negatively correlated with the time of action, and the inhibition rate(IR) increased in turn and positively correlated with the time of action of the low, middle and high dose groups of the gastric compound-containing plasma decreased. The W-SCR of G0/G1 phase cells was significantly increased, and was positively correlated with its dose in gastric cancer cells of different doses of gastric compound-containing plasma 72 h. Compared with the blank plasma group, the genes or protein expression of PTTG, c-Myc and hTERT in the 5-Fu group and the GFW group were down regulated. In dose-dependent manner after the gastric cancer cell in gastric compound-containing plasma 72 h.Conclusion: Gastric compound-containing plasma may down-regulate the expression genes of PTTG, c-Myc and hTERT to block the cycle and inhibit the proliferation of human gastric cancer BGC-823 cells to achieve antitumor effect.
