摘  要:目的探讨桂籽提取物对大鼠急性肺损伤的作用及其机制。方法将48只SD大鼠随机分为空白对照组、模型对照组、泼尼松组和桂籽粗提物大、中、小剂量组,每组8只。桂籽粗提物大、中、小剂量组分别给予桂籽粗提物400,200,100 mg·kg^-1,泼尼松组给予泼尼松3 mg·kg^-1,空白对照组和模型对照组给予等量0.9%氯化钠溶液。均连续灌胃10 d。模型对照组和各给药组于第10天灌胃结束1 h造模。24 h后经腹主动脉取血,开胸取肺。观察肺湿/干比(W/D)及肺组织水肿情况,苏木精-伊红(HE)染色观察肺部病理变化,测定各组髓过氧化物酶(MPO)、白细胞介素-6(IL-6)、IL-2、肿瘤坏死因子α(TNF-α)含量,Western blot法测定NF-κBp65及IκBα蛋白表达情况。结果与空白对照组比较,模型对照组大鼠肺部炎症细胞浸润明显,支气管壁明显增厚,水肿指数明显增大(W/D、LI值比较,P<0.01),血浆MPO、IL-6、IL-2、TNF-α含量明显升高,IκBα蛋白表达减少,NF-κBp65表达增加(P<0.05或P<0.01);与模型对照组比较,各给药组大鼠肺部炎症细胞浸润明显减少,支气管壁增厚明显改善,水肿指数明显减小(中、大剂量组LI值,P<0.05;中、大剂量组W/D值,泼尼松组W/D值、LI值,P<0.01)桂籽提取物可明显降低肺损伤大鼠MPO、IL-6、IL-2、TNF-α含量(P<0.05或P<0.01),提高IκBα蛋白表达,降低NF-κBp65表达(P<0.01)。结论桂籽提取物对大鼠急性肺损伤有一定保护作用,其机制一定程度上可能与上调IκBα以及激活NF-κBp65有关。Objective To explore the effect and mechanism of osmanthus fragrans seed extract on acute lung injury in rats. Methods Forty-eight SD rats were randomly divided into 6 groups(8 in each group). The rats of high,medium and low dose group were given 400,200,and 100 mg·kg^-1 osmanthus fragrans seed extract by gavage, respectively.In the positive control group,rats were given 3 mg·kg^-1 prednisone. The rats of blank control group and the model control group were given corresponding doses of 0.9% sodium chloride solution for 10 consecutive days. At 24 h and 12 h before the sampling,the model group,the positive control group and the drug-administered group were given 20% yeast suspension respectively,and the blank group was given the same amount of 0.9% sodium chloride solution. The lung wet/dry ratio and lung tissue edema were observed. The pathological changes of lung were observed by HE staining. The contents of MPO,IL-6,IL-2 and TNF-α in each group were determined. And the expression of NF-κB,p65 and IκBα protein was determined by Western blot. Results Compared with the blank control group,the inflammatory cells in the lungs of the model group were significantly infiltrated,the bronchial wall was thickened,the edema index was significantly increased(W/D、LI value,P<0.01),and the plasma levels of MPO,IL-6,IL-2 and TNF-α were significantly increased(P<0.01). The expression of IκBα protein decreased(P<0.01),and the expression of NF-κB p65 increased(P<0.01). Compared with the model group,the inflammatory cell infiltration of the lungs in the drug-administered group was significantly reduced, the bronchial wall thickening was significantly improved,and the edema index was significantly reduced(LI value in medium and large dose group, P<0.05;W/D value in medium and large dose groups, W/D value in positive dose group, LI value, P<0.01).
