HZ-835大孔树脂分离甘草黄酮的研究

摘　　要：通过静态吸附、解吸实验初步研究了8种大孔树脂对甘草黄酮的吸附与解吸性能,筛选出效果最佳的HZ-835型大孔树脂.以甘草黄酮的吸附率、解吸率为指标,通过动态吸附-解吸实验对5个影响因素进行考察,得出最佳的工艺条件为:上样料液的pH值为4.5、质量浓度2 mg/mL,7 g大孔树脂的最大上样量不超过40 mg;洗脱阶段,依次使用21.0 mL质量分数10%和10.5 mL质量分数20%的乙醇溶液对树脂柱进行洗涤,然后用31.5 mL质量分数70%的乙醇进行洗脱,吸附-解吸过程中的流速均为1 mL/min.大孔树脂纯化后收率为55.82%,纯度为15.76%,是纯化前的3.31倍;经液-液萃取后再用大孔树脂吸附,纯度能达到35.80%.Through static adsorption and desorption experiments,the adsorption and desorption properties of 8 kinds of macroporous resins on glycyrrhiza flavonoids were preliminarily studied,and the HZ-835 macroporous resin with the best effect was selected.Taking the adsorption rate and desorption rate of licorice flavonoids as indexes,five influencing factors were investigated through dynamic adsorption-desorption experiments,and the optimal process conditions were obtained as follows:the pH value of the feed liquid was 4.5,the mass concentration was 2 mg/mL,and the maximum loading amount of 7 g macroporous resin was not more than 40 mg;In the elution stage,the resin column was washed with 21.0 mL of 10%and 10.5 mL of 20%ethanol solution in turn,and then eluted with 31.5 mL of 70%ethanol in the adsorption-desorption process at a flow rate of 1 mL/min.After purification,the yield and purity of macroporous resin were 55.82%and 15.76%respectively,which was 3.31 times higher than that before purification.After liquid-liquid extraction and adsorption with macroporous resin,the purity can reach 35.80%.
