摘 要:目的探讨Ⅰ型单纯疱疹病毒(Herpes simplex virus type 1,HSV-1)感染人胚肺二倍体成纤维细胞(KMB-17)对长链非编码RNA(long non-coding RNA,lncRNA)的调控作用。方法用HSV-1感染KMB-17细胞,于感染后48h提取RNA进行高通量测序。测序获得的转录本用Cufflinks、CPC和CNCI软件进行lncRNA的鉴定和注释,并利用cuffdiff软件进行差异表达lncRNA的筛选。对筛选的差异表达lncRNA进行靶基因的预测及其GO和KEGG功能富集分析。结果共筛选到891个差异表达的lncRNA,其中上调515个,下调376个。GO和KEGG功能富集分析表明,HSV-1感染诱导表达差异的lncRNA靶基因主要富集在Toll样受体信号通路,RIG?I样受体信号通路,Jak-STAT信号通路、Hedgehog信号通路、趋化因子信号通路和细胞代谢相关信号通路上。结论 HSV-1感染KMB-17细胞后诱导细胞lncRNA差异表达并通过靶基因来影响细胞抗病毒免疫和细胞代谢等相关信号通路,调控细胞抗病毒的响应机制。Objective To investigate the regulatory effect of human embryo lung diploid fibroblasts(KMB-17)infected with herpes simplex virus type 1(HSV-1)on long non-coding RNAs(lncRNAs). Methods KMB-17 cells were infected with HSV-1.Total RNA was extracted at 48 hours post-infection for RNA-seq.The lncRNA transcripts were identified and annotated with the software cufflinks,CPC,and CNCI,and the differentially expressed lncRNAs were screened with the software cuffdiff.Target genes of lncRNAs were predicted,and the functions of target genes were analyzed using Go and KEGG enrichment analysis. Results A total of 891 differentially expressed lncRNAs were obtained,including 515 up-regulated and 376 down-regulated lncRNAs.The functional enrichment analysis of target genes of lncRNAs indicated that the target genes were mainly enriched in the Toll-like receptor signaling pathway,RIG-I-like receptor signaling pathway,JAK-STAT signaling pathway,hedgehog signaling pathway,chemokine signaling pathway,and a signaling pathway related to cell metabolism. Conclusion HSV-1 infection induced the differential expression of lncRNAs in KMB-17 cells,thus influencing the signaling pathways of antiviral immunity and cell metabolism through the target genes of dysregulated lncRNAs.
