摘 要:目的探讨低氧对蠕虫和细菌混合感染小鼠肠道黏膜免疫功能的影响。方法以十二指肠寄生鼠多形螺旋线虫和感染肠道鼠枸橼酸杆菌的小鼠为模型,分别在海拔2 260 m(大气压582mm Hg,PO2121.6mm Hg)和海拔5 000 m(大气压405mm Hg,PO284.7mm Hg)条件下建立单纯蠕虫感染及蠕虫和细菌混合感染模型,分别观察小鼠体重、小肠长度及十二指肠形态学变化;同时检测寄生在十二指肠的鼠多形螺旋线虫数目和各组小鼠血清IgE、IgG1抗体浓度。结果 (1)低氧组体重明显降低,低氧混合感染组降低尤为显著。(2)在低氧和蠕虫感染条件下小肠长度均增加。(3)低氧混合感染时,寄生在十二指肠的鼠多形螺旋线虫数目明显增多。(4)蠕虫感染时血清IgE、IgG1浓度均升高,但在低氧条件下小鼠IgE、IgG1抗体浓度有所降低。(5)十二指肠组织形态:低氧混合感染组有明显的黏膜破损和严重的上皮细胞脱落及炎细胞浸润。结论低氧会降低血清IgE、IgG1抗体浓度,影响肠道黏膜免疫功能;低氧可加重鼠多形螺旋线虫和肠道鼠枸橼酸杆菌混合感染导致的肠道病变。Objective To investigate the effect of hypoxia on the function of intestinal mucosal immune in mice co-infected with worms and bacteria.Methods Taking mice that were infected with Heligmosomoides Polygyrus(H.polygyrus,Hp)that parasitize in duodenum and Citrobacter rodentium(C.rodentium,Cr)as models.Establishing H.polygyrus infected groups and co-infected groups at 2260 m(582mmHg atmospheric pressure,PO2121.6mmHg)and 5000 m(405mmHg atmospheric pressure,PO284.7mmHg).In order to observe the weight changes,the length of small intestine and morphology of small intestine in mice.Then,checking the number of H.polygyrus and level of an-tibody in serum.Results①Body weight changes was significant in the hypoxic group,especially in the hypoxic co-infection group.②The length of the small intestine elongates during hypoxic conditions and parasitic infections.③The number of H.polygyrus increases in hypoxia co-infection group.④The serum levels of IgE and IgG1 were signif-icantly increased during parasite infection,but decreased under hypoxia.⑤Biopsy of duodenum showed significant mucosal damage,severe epithelium exfoliation and inflammatory cell infiltration in the hypoxia co-infected group.Conclusion The serum level of IgE and IgG1 was decreased under hypoxia,and affect the intestinal mucosal immu-nologic function;Hypoxia lead to severe intestinal lesions caused by concurrent infection of H.polygyrus and C.ro-dentium.
