摘 要:目的通过药效实验筛选复方黄芪方的工艺路线,并以正交实验进行优化,确定复方黄芪方的最佳提取工艺。方法通过高脂饲料喂养法建立高脂血症大鼠模型,以血清中三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、谷丙转氨酶(ALT)、谷草转氨酶(AST)水平及肝指数为指标,并结合肝组织病理切片的检测结果对复方黄芪方3条工艺路线进行筛选,得到最佳工艺路线;然后以黄芪甲苷和总黄酮含量为指标,通过正交实验优化该工艺路线的各提取工艺参数。结果药效实验结果表明,3条工艺路线中工艺路线一能显著改善大鼠血脂水平,最佳工艺为酒大黄单独粉碎成细粉;黄芪等其余药味水提,最佳提取工艺为加12倍量水,提取3次,每次1h。结论复方黄芪方能显著改善高脂血症大鼠的血脂水平,优选的制备工艺合理、可行,可为复方黄芪方后续开发提供依据。Objective To screen the preparation of compound Huangqi formula by pharmacodynamic experiment, and optimize by the orthogonal test. Methods A rat model of hyperlipidemia was established by high-fat diet. The serum triglyceride(TG), total cholesterol(TC), low-density lipoprotein cholesterol(LDL-C), alanine aminotransferase(ALT), aspartate aminotransferase(AST) levels and liver index were used as the indicators. Combined with the results of pathological sections of liver tissue, the 3 process routes of compound Huangqi formula were screened to decide the optimal route. The 3 process routes of auxiliary hypolipidemic formulas were screened. The content of astragaloside and total flavonoids was used as indicators, and orthogonal experiment was used to optimize parameters in the extraction process. Results Pharmacodynamic experiments showed that the process route 1 significantly improved the blood lipid level of rats. The best process was that the wine rhubarb was separately pulverized into fine powder, and other medicinal materials such as astragalus mixed with 12 times of water, 3 extractions, 1 hour each. Conclusion Compound Huangqi formula extract can significantly improve the blood lipid level in hyperlipidemia model rats. The preferred preparation process is stable and feasible, providing a basis for the subsequent development of compound Huangqi formula.
