摘 要:目的探讨添加白藜芦醇对于冻融卵巢组织异种移植后组织活力的改善作用。方法选取2014年6月行妇科手术的患者卵巢标本1例,使用玻璃化冷冻法保存,将卵巢组织解冻后移植到SCID鼠皮下,7 d后取出移植物,实验组采用白藜芦醇干预,对照组未添加白藜芦醇,比较移植物中卵泡数目,卵泡中凋亡发生率及微血管密度。结果实验组中异种移植后卵巢组织的卵泡数目与对照组相比显著升高,且卵泡凋亡的发生率显著低于对照组,差异有统计学意义(P<0.05);微血管密度计数提示实验组中异种移植后卵巢组织的微血管密度显著高于对照组,差异有统计学意义(P<0.05)。结论白藜芦醇添加在人类卵巢组织冷冻及异种移植体系中可改善移植物中卵泡存活率,但未来白藜芦醇作为抗氧化剂在人卵巢组织冷冻体系中的应用尚需进一步研究。Objective To determine the effect of resveratrol on ovarian tissue cryopreservation and transplantation.Methods One ovarian specimen from a patient undergoing gynecological surgery in June 2014 was selected and stored by vitrification,the ovary tissue was thawed and transplanted under the skin of SCID mice,the graft was removed 7 days later.The experimental group was treated with resveratrol,the control group did not add resveratrol,compared the number of follicles in the transplants,the incidence of apoptotic follicles and the microvascular density.Results The follicle number in experimental group was higher than in control group(P<0.05).The apoptosis rate in experimental group was lower than in control group.The microvascular density in experimental group was higher than in control group(P<0.05).Conclusion Resveratrol can increase the early viability of the follicles in ovarian tissues after xenotransplantation and this maybe a promising method to improve the protective effect of human ovarian tissue vitrification.
