摘  要:为了研究不同组织中α-L-岩藻糖苷酶1(α-L-fucosidase1,FUCA1)特异性表达的调节机制,运用实时荧光定量PCR(real-time quantitative PCR,qPCR)和蛋白免疫印迹(Western Blot)在mRNA水平和蛋白水平上分析了Fuca1基因在小鼠成体不同组织中的表达特点,还利用Clustal Omega分析了氨基酸序列在不同物种的同源性和系统进化树。qPCR检测结果显示,Fuca1基因在小鼠不同组织中存在表达差异,在睾丸中表达最高,其次是肾脏与卵巢;而Western Blot检测结果显示FUCA1主要在小鼠肾脏和卵巢高表达;对FUCA1氨基酸序列进行同源性分析,结果显示FUCA1保守性低。以上研究表明,FUCA1可能与小鼠的组织特异性表达调节有关,其在性腺中的具体表达机制还有待研究。The objective of this work is to explore the expression behavior ofα-L-fucosidase 1(FUCA1)and its regulatory mechanism in different tissues.To that end,real-time quantitative PCR(qPCR)and Western blot techniques were used to analyze mRNA and protein expressive characteristics of Fuca1 gene in different tissues in mouse.Also,the homology of FUCA1 amino acid sequences in different species was also analyzed by Clustal Omega.Our results of qPCR data show that the expression of Fuca1 gene was significantly different in mouse tissues,highest in testis,followed by ovary and kidney as the second highest.In addition,Western Blot results show that the FUCA1 was mainly expressed in mouse kidney and ovary.Our Clustal Omega alignment analysis results show that the amino acid sequence of FUCA1 had low conservation.These results indicate that FUCA1 may be related to the regulation of tissue-specific expression in mice,and the specific expression mechanism of FUCA1 in gonads remains to be resolved.
