摘 要:目的研究乙醛脱氢酶1 (ALDH1)细胞在宫颈癌Hela细胞系中的成瘤性及其干细胞特性。方法①利用流式细胞仪分选出宫颈癌Hela细胞系中ALDH1^+和ALDH1-细胞;②20例裸鼠随机分为ALDH1^+和ALDH-组,分别接种1×106个相应细胞;③裸鼠体内实验检测ALDH1^+和ALDH-细胞成瘤性。结果 ALDH1^+组细胞成瘤性为0. 428±0. 128,ALDH1-组细胞成瘤性为0. 141±0. 035,ALDH1^+组细胞成瘤性更高,两组比较差异有统计学意义(P<0. 05)结论 ALDH1^+细胞相比于ALDH-细胞具有更高的自我更新能力和致瘤性,可以作为干细胞标志物的应用,为宫颈癌治疗提供新的方向。Objective To detect the rate of tumor-forming of aldehyde dehydrogenase 1( ALDH1) cells in cervical cancer He La cell line,and analysis its stem cell characteristics. Methods ① by flow cytometry,ALDH1^+and ALDH1-cells in cervical cancer He La cell line were selected. ② 20 nude mice were randomly divided into ALDH1^+and ALDH-group,and every nude mouse were inoculated corresponding 1×106 cells,respectively. ③ in vivo tumorigenesis abilities were tested in NOD-SCID mice. Results The rate of tumor-forming of ALDH1^+ is 0. 428± 0. 128,and the rate of tumor-forming of ALDH1-is 0. 141 ± 0. 035. In comparision,ALDH1^+ cells had the higher tumor formation rate than ALDH1-cells( P<0. 05). Conclusion To compare with the ALDH1-cells,ALDH1^+ cells have the greater selfrenewal and tumorigenicity,which can be as a marker of cervical cancer stem cells,providing a new direction for the treatment of cervical cancer.
