摘  要:目的观察银杏叶提取物EGb761预处理对小鼠大脑微梗死(CMI)模型的影响。方法28只雄性C57BL/6J小鼠随机分为假手术组[n=6,PBS 100 mg(kg·d)]、磷酸缓冲液(PBS)组[n=11,PBS 100 mg/(kg·d)]和实验组[n=11,EGb761100 mg/(kg·d)]。PBS组和实验组小鼠均被双光子激光照射诱导建立CMI模型。HE染色计算微梗死体积、免疫荧光染色观察小鼠CMI区域神经元凋亡、小胶质细胞激活、星形胶质细胞过度活化及3-硝基酪氨酸(3-NT)沉积。结果假手术组未发现CMI病灶,大脑皮层组织的星形胶质细胞和小胶质细胞均呈现正常状态。与PBS组比较,实验组梗死体积、活化小胶质细胞、过度活化星形胶质细胞计数及3-NT沉积量减小(P<0.01)。结论EGb761预处理具有减少小鼠CMI区域神经元死亡,减小梗死体积的作用,机制可能与抑制小胶质细胞和星形胶质细胞活化,并降低硝化应激水平有关。Objective To explore the effect of Ginkgo Biloba extract EGb761 pretreatment for cerebral microinfarct(CMI) in mice. Methods Totally 28 male C57 BL/6 J mice were randomly divided into sham group(n=6, PBS 100 mg/kg per day), phosphate buffered saline(PBS) group(n=11, PBS 100 mg/kg per day), and experimental group(n=11, EGb761 100 mg/kg per day). CMI model was established by two-photon laser irradiation in experimental group and PBS group. The microinfarct size was caculated by HE staining. Apoptosis of neuron, activate microglia, overactive astrocyte, and deposition of 3-nitrotyrosine were observed by immunofluorescent staining. Results No CMI lesion was found in sham group. Astrocyte and microglia in cerebral cortex was normal. Compared with PBS group, infarct volume, counts of active microglia and overactive astrocyte, deposition of 3-nitrotyrosine decreased in experimental group(P<0.01). Conclusion EGb761 pretreatment can decrease neuron death and infarct volume in CMI area of mice, the mechanism may related with inhibiting over activation of microglia and astrocyte, and reducing nitrification stress level.
