摘 要:目的探究双和厚朴酚A(bishonokiol A,BHNKA)在大鼠体内的药代动力学过程。方法采用HPLC-FL法建立大鼠血浆中BHNKA的分析方法。8只大鼠灌胃BHNKA(40 mg/kg),测定不同时间点的血药浓度,绘制药时曲线,并用DAS 3.2软件计算相关药代动力学参数。结果大鼠血浆中BHNKA在0.98-250.00 ng/ml浓度范围内线性关系良好,平均提取回收率为86.8%-93.7%,日内、日间精密度均小于10%。主要药代动力学参数浓度-时间曲线下面积(AUC 0-∞)为(348.57±176.02)ng/(ml·h),消除半衰期(t 1/2)为(7.14±4.04)h,清除率(CL/F)为(0.14±0.06)L/(h·kg),达峰浓度(C max)为(84.97±32.58)ng/ml。结论本研究建立了操作简便、快速、灵敏度高的检测大鼠血浆中BHNKA的HPLC-FL方法。Objective To investigate the pharmacokinetics of bishonokiol A(BHNKA)in rats.Methods High-performance liquid chromatography-fluorescence detection(HPLC-FL)method was developed to determine the plasma concentration of BHNKA in rats.Eight rats were intragastrically administered with 40 mg/kg BHNKA to determine plasma concentrations at different time points and the data were analyzed with DAS 3.2 software.Results The linear relationship of BHNKA in plasma was well within the concentration range of 0.98-250.00 ng/ml.The recovery of BHNKA was 86.8%-93.7%.Intra-day precision and inter-day precision were both lower than 10%.The main pharmacokinetic parameter was(348.57±176.02)ng/(ml·h)for AUC 0-∞,(7.14±4.04)h for t 1/2,(0.14±0.06)L/(h·kg)for clearance,and(84.97±32.58)ng/ml for C max.Conclusion A simple,quick,and high sensitivity HPLC-FL method is established in this research,and it can be used for the analysis of pharmacokinetics of BHNKA in rat plasma.
