摘 要:目的:考察Menaquinone-7 (MK-7)对糖皮质激素诱导大鼠血管钙沉积的作用及机制。方法:大鼠连续注射地塞米松6周后,测定血清碱性磷酸酶(alkaline phosphatase,ALP)水平评定造模结果,将造模成功的大鼠随机分组给药。采用Von Kossa染色法检测血管壁的钙斑形成情况;放射免疫法测定血清骨桥蛋白(osteopontin,OPN)、护骨素(osteoprotegerin,OPG)、钙结合蛋白(calcium-binding protein,CaBP)及骨形态发生蛋白-7 (bone morphogenetic protein-7,BMP-7)水平;Western blot法测定血管生长停滞特异性基因产物6 (growth arrest specific gene 6,Gas 6)的蛋白水平;放射免疫法测定血栓素B2 (thromboxane B2,TXB2)、前列腺素E2 (prostaglandin E2,PGE2)及6-酮-前列腺素F1α(6-keto-prostaglandin F1α,6-keto-PGF1α)水平。结果:MK-7可抑制模型大鼠血管壁钙斑的形成,显著升高血清OPG、CaBP和BMP-7的水平以及血管Gas 6的蛋白水平,显著降低TXB2水平,升高PGE2及6-keto-PGF1α水平。结论:MK-7上调Gas 6等血管钙化抑制蛋白,抑制TXB2的形成,有利于抑制血管钙化。Objective: To investigate the effects of Menaquinone-7( MK-7) on glucocorticoid-induced vascular calcification in rats. Methods: Vascular calcification was established with dexamethasone in rats. Calcified plaque on the vascular wall was detected by Von Kossa staining. The levels of serum osteopontin( OPN),osteoprotegerin( OPG),calcium-binding protein( CaBP) and bone morphogenetic protein-7( BMP-7) were determined by radioimmunoassay( RIA) and the protein level of growth arrest specific gene 6( Gas 6) were measured with Western blot. Thromboxane B2( TXB2),prostaglandin E2( PGE2) and 6-keto-PGF1α were determined by RIA. Results: The formation of calcified plaque in vascular wall was inhibited by MK-7 in dexamethasone-treated rats. Compared with model group,the levels of serum OPG,CaBP,BMP-7 and the level of Gas 6 protein in blood vessel were significantly increased by MK-7. The level of TXB2 was decreased,while the levels of PGE2 and 6-keto-PGF1α were increased significantly by MK-7. Conclusion: MK-7 can inhibit vascular calcification by up-regulating the levels of serum OPG,CaBP,BMP-7 and the protein level of Gas 6 in blood vessel,and decreasing the formation of TBX2.
