摘  要:目的观察电针对不同阶段SAMP8小鼠学习记忆能力及海马区突触素(SYN)和突触后致密物-95(PSD-95)表达的影响,探讨电针治疗阿尔茨海默病的最佳时机。方法SAMP8小鼠48只随机分成模型组、3月龄电针组、6月龄电针组和9月龄电针组,每组12只,同龄正常老化SAMR1小鼠12只为对照组。电针组均选取“百会”“大椎”“肾俞”,电针频率为2 Hz,每日1次,8 d为1个疗程,每个疗程间隔2 d,共3个疗程。运用Morris水迷宫实验检测小鼠学习记忆能力,免疫组化和Western blot检测海马突触相关蛋白SYN和PSD-95的表达。结果与对照组比较,模型组小鼠逃避潜伏期明显延长,原平台象限停留时间明显缩短,跨越原平台次数明显减少,差异均有统计学意义(P<0.05,P<0.01),海马区SYN和PSD-95蛋白表达明显下降(P<0.01);与模型组比较,各电针组小鼠逃避潜伏期明显缩短,原平台象限停留时间明显延长,跨越原平台次数明显增加,差异均有统计学意义(P<0.05,P<0.01),海马区SYN和PSD-95蛋白表达均明显升高(P<0.01),且均以3月龄电针组最显著(P<0.05,P<0.01)。结论电针能提高SAMP8小鼠学习记忆能力,促进海马区SYN、PSD-95蛋白的表达,改善突触功能。Objective To observe the effects of electroacupuncture on learning and memory ability and expressions of synaptophysin(SYN)and postsynaptic density-95(PSD-95)in the hippocampus of SAMP8 mice at different stages;To explore the optimal time of electroacupuncture in the treatment of Alzheimer disease(AD).Methods Totally 48 SAMP8 mice were randomly divided into model group,3-month-old electroacupuncture group,6-month-old electroacupuncture group and 9-month-old electroacupuncture group,with 12 mice in each group.12 SAMR1 mice of the same age and normal aging were set as control group.In the electroacupuncture group,Baihui(GV20),Dazhui(GV14)and Shenshu(BL23)acupoints were selected.The electroacupuncture frequency was 2 Hz,once a day,and 8 days was a course of treatment,with a course interval of 2 days.There were 3 courses of treatment in total.Morris water maze experiment was used to test the learning and memory ability of mice in each group,and the expression of hippocampal synapse related proteins SYN and PSD-95 was measured by immunohistochemistry and Western blot.Results Compared with the control group,the escape latency of the model group was significantly prolonged,the residence time of the original platform quadrant was significantly shortened,and the number of crossing the original platform was significantly reduced,with statistical significance(P<0.05,P<0.01).The expression of SYN and PSD-95 protein in hippocampus of model group significantly decreased(P<0.01).Compared with the model group,the escape latency of the mice in each electroacupuncture group was significantly shortened,the residence time of the original platform quadrant was significantly prolonged,and the number of crossing the original platform significantly increased,with statistical significance(P<0.05,P<0.01).The expressions of SYN and PSD-95 protein in hippocampus of mice in each electroacupuncture group significantly increased(P<0.05,P<0.01),and were most significant in the 3-month-old electroacupuncture group(P<0.01).Conclusion Electroacupu
