摘  要:目的借助生物网络及模块化分析方法探索黄芪-丹参药对治疗慢性心力衰竭(CHF)的潜在作用靶点和信号,初步探索益气活血法治疗CHF可能的生物学机制。方法综合利用中药系统药理学数据库与分析平台(TCMSP)和中药分子机制的生物信息学分析工具(BATMAN-TCM)、DisgeNet和HPO数据库,分别获得药物有效成分、作用靶点及疾病相关治疗靶点,构建药对与疾病间的关键靶标网络,基于基因本体数据库GO和KEGG通路数据库确定靶标基因、蛋白质分子功能、细胞内定位及其所参与的生物学反应和通路。结果黄芪-丹参药对治疗CHF的作用机制可能与调节机体内激素的分泌、转运、代谢,机体循环系统调节,血液循环调节等有关,并可能作用于昼夜节律、固醇类激素合成、缺氧诱导因子、组氨酸代谢、肾素血管紧张素系统、基底转录因子等相关信号通路。结论黄芪-丹参药对可能通过调节相关物质基础发挥治疗CHF作用,“昼夜节律调控”可能成为益气活血法治疗CHF新的思路。Objective To explore the potential targets and signals of Astragali Radix-Salviae Miltiorrhizae et Rhizoma medicinal pair for the treatment of chronic heart failure(CHF)by means of biological networks and modular analysis methods;To preliminarily explore the possible biological mechanism of Yiqi Huoxue therapy for CHF.Methods The TCMSP and BATMAN-TCM,DisgeNet and HPO databases were used to obtain the active ingredients,targets and disease-related therapeutic targets respectively.The key target network between medicinal pairs and diseases was constructed.Target genes,protein molecular functions,intracellular localization,and the biological responses and pathways involved were determined based on GO and KEGG pathway database.Results The mechanism of Astragali Radix-Salviae Miltiorrhizae et Rhizoma medicinal pair for the treatment of CHF may be related to the regulation of hormone secretion,transport,metabolism,regulation of circulatory system,regulation of blood circulation,etc.,and may act on circadian rhythm,steroid hormone synthesis,hypoxia induction factors,histidine metabolism,renin angiotensin system,basal transcription factors and other related signaling pathways.Conclusion Astragali Radix-Salviae Miltiorrhizae et Rhizoma medicinal pair may play a role in the treatment of CHF by regulating the related substances.The regulation of circadian rhythm may become a new idea for the treatment of CHF by Yiqi Huoxue therapy.
