摘 要:目的建立超高效液相色谱法同时测定盐肤木中8种成分含量方法,为盐肤木药材的质量控制提供依据。方法采用ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7μm),流动相为乙腈-0.1%甲酸水梯度洗脱,流速0.3 mL/min,柱温35℃,检测波长275 nm。结果没食子酸、苔黑酚葡萄糖苷、没食子酸甲酯、3,5-二羟基甲苯、鞣花酸、漆黄素、五没食子酰葡萄糖和根皮苷的线性范围分别为23.31~1165.5 ng(r=0.9992)、27.02~1351.5 ng(r=0.9991)、6.30~315.0 ng(r=0.9992)、12.36~618.0 ng(r=0.9992)、17.14~857.5 ng(r=0.9996)、9.24~461.5 ng(r=0.9998)、25.29~1264.5 ng(r=0.9996)和6.18~309.0 ng(r=0.9993),平均加样回收率为96.77%~99.41%(RSD为2.23%~3.24%)。盐肤木样品经聚类分析和主成分分析可分为两类,且没食子酸、五没食子酰葡萄糖、没食子酸甲酯和漆黄素等成分在两类中的分布存在差异。结论本研究建立的方法可为完善盐肤木药材的质量标准和加强质量控制提供依据。Objective To establish a UPLC method to simultaneously determine the contents of eight chemical compositions in the root of Rhus chinensis Mill.;To provide references for quality control of Rhus chinensis Mill.Methods The UPLC method was performed on an ACQUITY UPLC BEH C18 column(2.1 mm×100 mm,1.7μm)with a gradient elution using acetonitrile and 0.1%formic acid.The flow rate was 0.3 mL/min;the column temperature was 35℃;the detection wavelength was set at 275 nm.Results Gallic acid,orcinol glucoside,methyl gallate,3,5-dihydroxytoluene,ellagic acid,fisetin,pentagalloylglucose and phlorizin showed good linear relationship within the range of 23.31–1165.5 ng(r=0.9992),27.02–1351.5 ng(r=0.9991),6.30–315.0 ng(r=0.9992),12.36–618.0 ng(r=0.9992),17.14–857.5 ng(r=0.9996),9.24–461.5 ng(r=0.9998),25.29–1264.5 ng(r=0.9996),and 6.18–309.0 ng(r=0.9993),respectively.The overall recoveries were ranged from 96.77%to 99.41%with the RSD range of 2.23%–3.24%.After cluster analysis and principal components analysis,the collected samples of Rhus chinensis Mill.could be divided into two types of patterns,and there are differences in the distribution of compositions in the two types,such as gallic acid,pentagalloylglucose,methyl gallate and fisetin.Conclusion The method can provide a basis for improving the quality standard and quality control of Rhus chinensis Mill.
