摘 要:[目的]研究越鞠丸对功能性消化不良(functional dyspepsia,FD)大鼠胃肠功能的改善作用,并探讨其可能的作用机制。[方法]48只SPF级健康雄性SD大鼠,称重后随机分为6组:对照组、模型组、莫沙必利组及越鞠丸低、中、高剂量组,每组各8只。对照组不予特殊处理,其他各组采用郭氏夹尾法建立FD大鼠模型,通过大鼠体重、进食及饮水量、胃排空率,胃窦黏膜形态评估造模情况。对照组、模型组予0.9%氯化钠溶液2mL/次,莫沙必利组予0.3mg/200g,越鞠丸低、中、高剂量组分别予0.9g/200g、1.8g/200g、3.6g/200g中药浓缩液连续灌胃7d,1次/d。比较各组大鼠造模、给药前后体重、进食及饮水量改变情况;计算给药后大鼠胃排空率;采用免疫组化及酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法检测胃窦组织5-羟色胺(5-hydroxytryptamine,5-HT)的含量和血清血管活性肠肽(vasoactive intestinal peptide,VIP)的水平。[结果]与对照组比较,模型组大鼠的体重、进食及饮水量均显著下降,差异有统计学意义(P<0.05),说明FD大鼠模型造模成功。与对照组比较,模型组大鼠胃排空率下降(P<0.05);与模型组比较,莫沙必利组、越鞠丸中、高剂量组大鼠胃排空率均增加,差异具有统计学意义(P<0.05)。与模型组比较,莫沙必利组和越鞠丸中、高剂量组大鼠胃窦组织5-HT含量升高、血清VIP水平减低,差异有统计学意义(P<0.05)。与莫沙必利组比较,越鞠丸高剂量组胃窦组织5-HT含量差异无统计学意义(P>0.05),越鞠丸中、高剂量组血清VIP水平差异无统计学意义(P>0.05)。[结论]越鞠丸能够增加FD大鼠的胃肠动力,其作用机制可能是通过增加胃窦组织5-HT表达、下调血清VIP水平来影响胃肠功能。[Objective]To investigate the influence-of Yueju Pill on the improvement of gastrointestinal function in rats with functional dyspepsia(FD)and to explore its possible mechanism.[Methods]Forty-eight specific pathogen free(SPF)healthy male SD rats were randomly divided into six group after being weighed,control group,model group,Mosapride group,Yueju Pill low dose group,Yueju Pill medium dose group and Yueju Pill high dose group,with 8 in each group.Rats in control group were not treated,and the GUO’s nipped tail method was applied to establish the FD rat model in other groups.Rats in control group and model group were given 0.9%sodium chloride solution 2 mL/time,Mosapride group was given 0.3 mg/200 g,and Yueju Pill low,medium and high dose groups were given 0.9 g/200 g,1.8 g/200 g and 3.6 g/200 g Chinese medicine concentrate respectively,and each group was administrated intragastrically once a day for 7 days.Subsequently,the changes of body weight,food and water consumption before and after model establishment and treatment were compared,and the gastric emptying rate after given drug was calculated.Immunohistochemistry and enzyme-linked immunosorbent assay(ELISA)were used to detect 5-hydroxytryptamine(5-HT)contant in gastric antrum and serum vasoactive intestinal peptide(VIP)level of serum.[Results]Compared with control group,the body weight,food and water consumption decreased significantly in model group(P<0.05),indicating that the FD rat model was successfully modeled.Compared with control group,the gastric emptying rate of model group decreased(P<0.05).Compared with model group,the gastric emptying rate of Mosapride group,Yueju Pill medium and high dose groups increased,and the difference was statistically significant(P<0.05).Compared with model group,5-HT content in gastric antrum increased,and serum VIP level decreased in Mosapride group,Yueju Pill medium and high dose groups,the difference was statistically significant(P<0.05).Compared with Mosapride group,there was no significant difference of 5-HT coate
