摘 要:目的观察DA、5-HT及MAO在甲基苯丙胺和氯胺酮联合应用致大鼠成瘾中的表达变化及其与依赖大鼠产生CPP效应的关系。方法随机将36只大鼠分为六组:正常对照组,甲基苯丙胺组(2 mg/kg),氯胺酮低剂量(15 mg/kg)组,氯胺酮中剂量(30 mg/kg)组,甲基苯丙胺+氯胺酮低剂量(2 mg/kg+15 mg/kg)组,甲基苯丙胺+氯胺酮中剂量(2 mg/kg+30 mg/kg)组,每组每天给予相应的药物,连续10 d,建立大鼠位置偏爱模型;ELISA检测DA、5-HT及MAO的表达。结果与正常对照组比较,给药组大鼠在伴药箱中逗留的时间均明显增多(P<0.01),成功建立位置偏爱模型。ELISA检测结果显示,在大鼠前额叶皮质、伏隔核、中脑腹侧被盖区中DA、5-HT及MAO的表达趋势基本相同;与正常对照组相比,MA组表达增强(P<0.01);与MA组相比,联用组的DA、5-HT及MAO表达均呈一致性降低(P<0.01)。结论DA、5-HT及MAO表达水平降低与甲基苯丙胺和氯胺酮联合给药的依赖大鼠CPP效应没有显著提升有关。Objective To observe the expression changes of DA,5-HT and MAO in the addiction induced by methamphetamine and ketamine abuse and their relationship with conditioned place preference(CPP)effect in dependent rats.Methods A total of 36 rats were randomly divided into 6 groups:normal control group,methamphetamine group(2 mg/kg),low-dose ketamine group(15 mg/kg),high-dose ketamine group(30 mg/kg),methamphetamine+ketamine low dose group(2 m/kg+15 mg/kg),methamphetamine+ketamine high dose group(2 mg/kg+30 mg/kg).Each group was given the corresponding drugs every day for 10 days to establish the rat model of CPP.ELISA method was applied to detect DA,5-HT and MAO expression.Results Compared with the normal control group,the rats in the administration groups showed significantly extended stay in the drug-paired compartment(P<0.01)and the CPP was established.The results of the ELISA showed the expression trends of DA,5-HT,and MAO in the prefrontal cortex,nucleus accumbens,and the ventral tegmental area(VTA)were basically the same.Compared with that of the normal control group,the expression of the methamphetamine group was enhanced(P<0.01).Compared with those in the MA group,the expressions of DA,5-HT and MAO in the combined group showed consistent decrease(P<0.01).Conclusion The decreased expression of DA,5-HT and MAO is associated with no significant increase of CPP in dependent rats treated with methamphetamine and ketamine.
