摘 要:【目的】观察白藜芦醇对代谢综合征(MS)大鼠肠道菌群的影响。【方法】采用高脂高盐高糖饮食诱导大鼠MS模型。将造模成功的MS大鼠随机分为模型组、白藜芦醇组和二甲双胍组,并另设正常组予普通饲料喂养,后连续灌胃对应给药或饮用水4周。末次灌胃后,取大鼠粪便进行培养,观察肠道主要菌群数量的变化;酶联免疫吸附分析(ELISA)检测血清肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素6(IL-6)水平;逆转录聚合酶链反应(RT-PCR)法检测肝组织禁食诱导脂肪因子(Fiaf)、脂蛋白脂酶(LPL)、乙酰辅酶A羧化酶α(ACCα)、脂肪酸合成酶(Fas)mRNA表达水平。【结果】与模型组比较,白藜芦醇组大鼠粪便大肠杆菌和链球菌数量显著降低,双歧杆菌、乳酸杆菌数量显著增加(P<0.05或P<0.01),血清TNF-α、IL-1β和IL-6含量明显降低,肝组织Fiaf mRNA表达量明显升高,LPL、ACCα和Fas mRNA表达量下降(P<0.05或P<0.01)。【结论】白藜芦醇能明显减轻MS大鼠炎症反应及调控脂代谢相关靶基因的表达,其机制可能与调节肠道微生态平衡,促进益生菌的增长有关。Objective To observe the effects of resveratrol on intestinal flora in rats with metabolic syndrome(MS).Methods MS rat model was established by feeding high-fat,high-saline and high-glucose diet.The successfulMS model rats were randomly divided into model group,resveratrol group and metformin group.Additionally,thenormal group was set up and fed with common diet.And then the various groups were respectively given intragastricadministration of corresponding drug or distilled water for 4 weeks.After the last administration,the faeces wascultured for observing the changes in intestinal flora,the levels of tumor necrosis factorα(TNF-α),interleukin1β(IL-1β)and interleukin 6(IL-6)were detected by enzyme-linked immuno sorbent assay(ELISA),and them RNA expression levels of fasting-induced adipose factor(Fiaf),lipoprotein Lipase(LPL),acetyl-Co Acarboxylaseα(ACCα)and fatty acid synthetase(Fas)were determined by reverse transcription-polymerase chainreaction(RT-PCR).Results Compared with the model group,the number of pathogenic E.coli and Streptococcus was decreased in faeces of the resveratrol group,the number of Lactobacilli and Bifidobacterium was elevated(P<0.05 or P<0.01),the serum levels of TNF-α,IL-1βand IL-6 were decreased,and the m RNA expression levelof Fiaf was increased and the m RNA expression levels of LPL,ACCαand Fas were decreased in liver tissues(P<0.05 or P<0.01).Conclusion Resveratrol has significant effects on relieving the inflammatory reaction,andregulating and controling the expression of adipose genes related with lipid metabolism in MS rats,whosemechanism may be associated with adjusting intestinal microecological balance and promoting the growth of probiotics.
