摘 要:目的优化槐米的提取工艺以提高芦丁的提取率。方法在碱提酸沉的基础上,选用电热套加热(130℃,微沸)回流30min,再超声辅助(70℃,40kHz,200W)提取30min的方法提取芦丁。对影响芦丁提取率的因素如pH值、料液比和静置时间进行了考察。应用UV法和HPLC法对自提芦丁进行了检识。结果最优提取工艺为碱液溶解时pH值为8,料液比为1∶20(g/mL),酸沉淀后静置时间为24h。UV法和HPLC法检识结果表明此法自提的芦丁较纯。结论优化提取工艺可提高槐米中芦丁的提取率。Objective To optimize the extraction technology of sophora japonica to improve the extraction rate of rutin.Methods On the basis of alkali extraction and acid precipitation,rutin was extracted by electric heating(130℃,slightly boiling)for reflux for 30 min,and ultrasonic assisted extraction(70℃,40kHz,200W)for 30 min.Factors influencing the extraction rate of rutin such as pH value,solid-liquid ratio and standing time were investigated.UV method and HPLC method were used to detect rutin.Results The pH value of dissolved lye in the optimal extraction process was 8,the ratio of material to liquid was 1∶20(g/mL),and the acid precipitation time was 24 hours.The results of UV and HPLC showed that the rutin extracted by this method was relatively pure.Conclusion Optimized extraction technology can improve the extraction rate of rutin from sophora japonica.
