摘　　要：目的通过检测孕早期母体血清Kisspeptin水平和绒毛组织的表达,拟探讨其在妊娠中的作用。方法采用免疫组织化学方法分析正常妊娠组(20例)和自然流产组(20例)绒毛组织中Kisspeptin的表达水平;采用酶联免疫吸附法(ELISA)检测孕5~10周妇女(40例)血清Kisspeptin浓度。结果①免疫组织化学分析显示,Kisspeptin在正常妊娠组[灰度值(170.3±7.134)]的表达显著高于自然流产组[灰度值(198.4±5.699))。②ELISA法结果显示,正常妊娠组Kisspeptin在血清中的浓度随孕周增大而增高,自然流产组Kisspeptin在血清中的浓度随孕周逐渐降低。结论Kisspeptin在孕早期胚胎着床及发育过程中起着重要作用,可作为一种预测妊娠结局的指标。Objective To explore the effect of Kisspeptin in early stages of pregnancy by detecting it’s expression levels in the serum and villus tissue.Methods Immunohistochemical method was used to analyze the expression of kisspeptin in chorionic villi of normal pregnancy group(n=20)and spontaneous abortion group(n=20).The serum kisspeptin concentration of 40 pregnant women at 10 weeks of gestation was detected by enzyme-linked immunosorbent assay(ELISA).Results①Normal pregnant women had significantly higher villus tissue kisspeptin levels(170.3±7.134)than abortion women(198.4±5.699)(P<0.001).②The results of ELISA showed that the serum concentration of Kisspeptin in normal pregnancy group increased with the increase of gestational age,while the serum concentration of kisspeptin in spontaneous abortion group decreased gradually with gestational age.Conclusion Kisspeptin plays an important role in embryo implantation and development in early pregnancy,and can be used as an indicator to predictpregnancy outcome.
