摘 要:为筛选响应月季花朵衰老的RhRRs基因以及分析其表达特性,采用RT-PCR,RACE PCR以及MEGA等生物技术方法,检测了RhRRs基因在月季花朵不同开放阶段的表达谱,克隆了Rh39694基因全长,开展了进化树分析.结果表明,RU39694与RU12149的表达谱受月季花朵衰老显著抑制;其中RU39694基因全长1292 bp,开放阅读框(ORF)为738 bp,编码245个氨基酸,含有A类RRs蛋白特有的DDK基序,蛋白分子式为C1176 H1912 N 324 O 401 S12,分子量为27.39 kDa,等电点(pI)为4.88;RU39694蛋白与拟南芥ARR8,ARR9亲缘关系最近,被命名为RhRR9;此外,RhRR9的表达谱受外源细胞分裂素(6-BA)处理显著诱导,而被脱落酸(ABA)与乙烯(C 2H 4)处理显著抑制,推测可能在植物激素(细胞分裂素、脱落酸、乙烯)拮抗调控月季花朵衰老中发挥重要功能.In order to screen RhRRs in response to rose flower senescence and to analyze their expression patterns,RT-PCR was used to monitor expression profiles of RhRRs in rose flower at different flowering stages and after different plant growth regulators treatments.Addtionally,RACE-PCR and MEGA were performed to clone the full length of Rh39694 and to analyze phylogenetic tree,respectively.The results showed that among four RhRRs,the transcripts of both RU39694 and RU12149 were significantly repressed during rose flower senescence.The full length of RU39694 was1292 bp,including an open reading frame of 738 bp.It encoded a protein of 245 amino acids with a typical DDK motif,which was conserved in Type-A RRs family.The formula of this protein was C1176 H1912 N 324 O 401 S 12 with a molecular weight of 27.39 kDa and a theoretical pI of 4.88.Phylogenetic tree analysis suggested that RU39694 shared high similarity with Arabidopsis ARR8 and ARR9,and was designated RhRR9.Additionally,the expression of RhRR9 was significantly induced by 6-BA treatment,while was strongly repressed by ABA or ethylene treatments.Taken together,these results supported an hypothesis that RhRR9 may play a critical role in regulation of rose flower senescence via antagonism of cytokinin to ABA and ethylene.
