摘 要:目的:使用聚乳酸-羟基乙酸共聚物(PLGA)携带包载三七皂苷Rb1制备纳米微球并优化实验条件,寻求包封率及平均粒径的最优制备条件。方法:制备纳米微球使用复乳-溶剂挥发法,使用高效液相色谱仪测定包封率,激光粒度分析仪测定平均粒径。使用SPSS 13.0统计学软件,对影响包封率及平均粒径的因素分别进行五因素四水平正交实验,分析包封率及粒径制备的最优条件。结果:PLGA浓度10 mg/mL,O︰W1为10︰3,W2︰W1/O为2︰1,第1次超声乳化时间10 s,第2次超声乳化时间90 s的条件下制备的微球包封率最佳。对于粒径来说,每个因素最佳的水平分别为PLGA浓度20 mg/mL,O︰W1为1︰5,W2︰W1/O为2︰1,第1次超声乳化时间10 s,第2次超声乳化时间120 s。结论:高分子有机化合物PLGA可作为为外壳材料包被携带三七皂苷Rb1,通过实验优化可获得其包封率及粒径制备的最优条件。Objective:To prepare nanospheres of panax notoginseng saponin Rb1 with PLGA and optimize the experimental conditions,and to find the optimal preparation conditions of encapsulation rateand average particle size.Method:PLGA loaded panax notoginseng saponin Rb1 nanoparticles were prepared by double emulsion solvent evaporation method.The encapsulation rate was determined by high performance liquid chromatograph and the average particle size was determined by laser particle size analyzer.Five factors and four levels of orthogonal experiments were carried out on the factors affecting the encapsulation rate and the average particle size by SPSS 13.0 statistical software.The optimal conditions of encapsulation rate and particle size preparation were analyzed.Result:When PLGA concentration was 10 mg/mL,O︰W1 was 10︰3,W2︰W1/O was 2︰1,the first time of phacoemulsification was 10 s,and the second time of phacoemulsification was 90 s,the encapsulation efficiency of microspheres was the best.PLGA concentration was 20 mg/mL,O︰W1 was 1︰5,W2︰W1/O was 2︰1,the first time of phacoemulsification was 10 s,the second time of phacoemulsification was 120 s for the best preparation factor of particle size.Conclusion:The polymeric organic compound PLGA can be used as the shell material to carry panax notoginseng saponin Rb1.The optimal conditions of encapsulation rate and particle size preparation can be obtained by experimental optimization.
