摘  要:为探明黄瓜磷脂酶Dα(CsPLDα)和促分裂原活化蛋白激酶(CsNMAPK)基因对短时间Na Cl胁迫(0 h, 0.5h, 1 h, 3 h, 5 h and 10 h)的响应,以‘新泰密刺’试材,研究了100 m M Na Cl不同时间处理下黄瓜幼苗叶片中CsPLDα和CsNMAPK表达和相关信号分子的变化。结果表明:随处理时间延长,黄瓜幼苗叶片中CsPLDα和CsNMAPK表达量呈先升高后降低又略有升高趋势,CsPLDα表达量在处理1 h后达最大值,而CsNMAPK表达量在处理3 h后达最大值;两者的蛋白表达量亦分别在处理1 h和3 h达最大值;信号分子磷脂酸(PA)含量亦呈先升高后降低趋势,在处理3 h后达最大值;过氧化氢和超氧阴离子含量随处理时间延长呈逐渐增加趋势;超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物歧化酶(POD)和抗坏血酸过氧化物酶(APX)活性呈先降低后升高又降低趋势,均在处理0.5 h后显著降低,SOD活性在处理3 h后达较高值,但与处理0 h无显著差异;CAT、POD和APX活性均在处理5 h后较高,显著高于0 h。综上可知,黄瓜中CsPLDα和CsNMAPK表达对Na Cl短时间胁迫响应存在时间差,且CsPLDα响应早于CsNMAPK。In order to find out the response of cucumber phospholipase D α(CsPLD α) and mitogen activated protein kinase(CsNMAPK) gene to short time Na Cl stress(0 h, 0.5 h, 1 h, 3 h, 5 h and 10 h), the changes of CsPLD α and CsNMAPK expression and related signal molecules in cucumber seedling leaves treated with 100 m M Na Cl at different time were studied with the test material of "xintaimici". The results showed that with the prolongation of processing time, the expression of CsPLDα and CsNMAPK in cucumber seedling leaves increased first, then decreased and then slightly increased.The expression levels of CsPLDα reached the maximum value after 1 h of treatment, while the expression levels of CsNMAPK reached the maximum value after 3 h treatment. The protein expression of them reached maximum at 1 h and 3 h,respectively. The content of signal molecule phosphatidic acid(PA) increased at first and then decreased, and reached the maximum value after 3 h treatment. The contents of hydrogen peroxide and superoxide anion increased gradually with the prolongation of treatment time. The activities of superoxide dismutase(SOD), catalase(CAT), peroxidase(POD) and ascorbic acid peroxidase(APX) decreased first, then increased, and then decreased, all of them decreased significantly after0.5 h of treatment, and SOD activity reached a higher value after 3 h treatment, but there was no significant difference with the treatment for 0 h. The activities of CAT, POD and APX were all higher after 5 h of treatment, significantly higher than that of 0 h. In conclusion, there was a time difference between the expression of CsPLD α and CsNMAPK in cucumber to short time Na Cl stress, and the response of CsPLD α was earlier than that of CsNMAPK.
