摘 要:为探讨玫瑰香系葡萄种质资源间的遗传关系,利用SRAP标记技术对39个玫瑰香系葡萄品种进行了遗传多样性分析,并构建其DNA指纹图谱。筛选出的14对引物共扩增出139条带,其中多态性条带110条,多态性比率为79.14%。14对引物的多态性信息含量的平均值为0.862 8,观测等位基因数的平均值为1.791 4,有效等位基因数的平均值为1.379 1,Nei's基因多样性指数的平均值为0.226 9,Shannon's信息指数的平均值为0.348 8。聚类结果显示,39个玫瑰香系葡萄品种间的遗传相似系数在0.604 3~0.978 4之间,变幅为0.374 1。上述结果表明:39个玫瑰香系葡萄品种间的具有较丰富的遗传多样性水平。在遗传相似系数为0.76处可将39个玫瑰香系葡萄品种分为4组,其中,第Ⅰ组有8个欧美杂交种,15个欧亚种;第Ⅱ组有6个欧美杂交种和1个欧亚种;第Ⅲ组7个品种均为欧亚种;第Ⅳ组2个品种均为欧美杂交种。利用8对引物扩增的19个多态性位点构建了39个玫瑰香系葡萄品种的DNA指纹图谱,该图谱可为玫瑰香系葡萄品鉴定提供科学依据。In order to explore the genetic relationship among the germplasm resources of Muscat Hamburg grapevine series,the genetic diversity of 39 cultivars belonging to Muscat Hamburg grapevine series was analyzed by SRAP marker,and their DNA fingerprints were constructed.A total of 139 bands were amplified from 14 pairs of primers,of which 110 were polymorphic bands with a polymorphic ratio of 79.14%.The average of the polymorphism information content(PIC)of 14 pairs of primers was 0.8628,the average number of observed alleles(Na)was 1.7914,the average number of effective alleles(Ne)was 1.3791,the average value of Nei’s gene diversity index(H)was 0.2269,and the average value of the index(Ⅰ)was 0.3488.The results of clustering showed that the genetic similarity coefficient among 39 cultivars ranged from 0.6043 to 0.9784,with a variation of 0.3741.The above results indicate that the genetic diversity of 39 cultivars was higher.39 cultivars were divided into 4 groups with a genetic similarity coefficient of 0.76.Among them,groupⅠhas 8 Euro-American hybrids species and 15 V.vinifera;GroupⅡhas 6 Euro-American hybrids species and 1 V.vinifera;GroupⅢ7 cultivars were V.vinifera;GroupⅣ2 cultivars were Euro-American hybrids species.The DNA fingerprints of 39 cultivars were constructed by using 19 polymorphic loci amplified by 8 pairs of primers.This map could provide scientific basis for the identification of Muscat Hamburg grapevine series.
