摘  要:为探究合适的解离液类型用于流式细胞术检测芜菁倍性与相对DNA含量,明确新疆地区主栽芜菁品种的DNA含量和倍性,同时检测游离小孢子培养所获得的再生植株倍性。本研究选取‘阿恰芜菁’、‘温州盘菜’及小孢子培养所获得的再生植株群体为试验材料,在常用的10种解离液中筛选适合芜菁的解离液类型。LB01适合作为流式细胞术检测芜菁样品的最适解离液。以四倍体紫花苜蓿‘金皇后’为外标,检测芜菁的DNA含量及倍性后发现‘温州盘菜’的DNA含量为0.657 pg,倍性为二倍体。‘阿恰芜菁’的DNA含量为0.723 pg,倍性为二倍体。小孢子再生群体Ⅰ的DNA含量为0.626 pg,倍性为二倍体。小孢子再生群体Ⅱ的DNA含量为2.225 pg,倍性为四倍体。本研究为流式细胞术检测芜菁倍性和DNA含量的方法提供了数据参考,也将为芜菁群体变异、种质资源评价和新品种选育等研究提供基础。To explore the type of dissociation solution suitable for detecting the ploidy and relative DNA content of turnip by flow cytometry,so as to clarify the DNA content and ploidy of main varieties of turnip in Xinjiang.The regenerated plant populations obtained from’Aqiawujing’,’Wenzhoupancai’and microspore culture were selected as experimental materials,and the suitable types of dissociation solution for turnip were screened in 10 common dissociation solutions.LB01 was suitable for flow cytometry to detect turnip samples.Taking the tetraploid alfalfa’Golden Queen’as the external standard,the DNA content and ploidy of turnip were detected.It was found that the DNA content of’Wenzhoupancai’was 0.657 pg and the ploidy was diploid.The DNA content of’Aqiawujing’was 0.723 pg and its ploidy was diploid.The DNA content of microspore regeneration populationⅠwas 0.626 pg and its ploidy was diploid.The DNA content of microspore regeneration populationⅡwas 2.225 pg and its ploidy was tetraploid.This study provides a data reference for flow cytometry to detect ploidy and DNA content of turnip,and also provides a basis for population variation,germplasm resource evaluation and new variety breeding of turnip.
