红系祖细胞及其亚型在肺结核患者外周血的比例及作用研究

摘  要:目的探讨红系祖细胞及其亚型在肺结核患者外周血的比例及作用。方法收集2016年1月至2018年12月来我院就诊的肺结核初治患者,HE染色、抗酸染色和免疫组化检测肺组织的结核病变;流式细胞术检测红系祖细胞及其亚型、T细胞和CD8^+T细胞比例;采用Transwell小室进行细胞共培养实验。结果结核组肺组织呈慢性肉芽肿性炎改变,CD68^+巨噬细胞浸润,抗酸染色可检测到阳性的结核分枝杆菌;对照组和结核组之间红系祖细胞和CD45-红系祖细胞比例均没有显著的统计学差异,结核组CD45^+红系祖细胞比例显著的高于对照组(t=6.801,P<0.0001);对照组和结核组CD45^+红系祖细胞ROS的平均荧光强度均显著的高于CD45^-红系祖细胞(t=5.452,P=0.0003;t=5.083,P=0.0005);共培养实验结果显示CD45^+红系祖细胞能够通过ROS通路引起T细胞和CD8^+T细胞比例下降,CD45^-红系祖细胞则没有显著的作用。结论CD45^+红系祖细胞在肺结核患者外周血中显著升高,CD45^+红系祖细胞能够通过ROS通路抑制T细胞和CD8^+T细胞免疫。This study was preformed to investigate the proportion and role of erythroid progenitor cells and their subtypes in peripheral blood of patients with pulmonary tuberculosis.The patients with pulmonary tuberculosis were recruited from our hospital from January 2016 to December 2018.HE staining,acid-fast staining and immunohistochemistry were used to detect the tuberculous changes in lung tissue;flow cytometry was used to detect the proportion of erythroid progenitor cells and their subtypes,T cells,and CD8^+T cells.Transwell cell chamber was used for cell co-culture experiment.The lung tissue of tuberculosis group showed chronic granulomatous inflammation,CD68^+macrophage infiltration,and positive Mycobacterium tuberculosis confirmed by acid-fast staining.However,there was no significant difference in the proportion of erythroid progenitor cells and CD45^-erythroid progenitor cells between the control group and the tuberculosis group,while the proportion of CD45^+erythroid progenitor cells in the tuberculosis group was significantly higher than that in the control group(t=6.801,P<0.0001).The MFI of CD45^+erythroid progenitor cells was significantly higher than that of CD45_erythroid progenitor cells in both control group and tuberculosis group(t=5.452,P=0.0003;t=5.083,P=0.0005).The coculture experiment showed that CD45^+erythroid progenitor cells could decrease the proportion of T cells and CD8^+T cells through ROS pathway,but CD45^-erythroid progenitor cells had no significant effect.In conclusion,CD45^+erythroid progenitor cells are significantly increased in the peripheral blood of patients with pulmonary tuberculosis,and can inhibit T cell and CD8^+T cell immunity through ROS pathway.
