摘 要:为了提高草鱼皮的附加值,采用酶法制备胶原蛋白抗氧化肽.以DPPH自由基清除率为指标,从5种蛋白酶中筛选出碱性蛋白酶为最佳蛋白酶.通过Plackett-Burman试验设计、最陡爬坡试验和Box-Behnken中心组合试验设计优化出草鱼皮胶原蛋白的最优酶解条件为:酶解时间3.0h、酶解温度46.5℃、加酶量7025.2 U/g;在此条件下,草鱼皮胶原蛋白酶解产物对DPPH自由基清除率可达78.3%,为理论值的100.4%.超声预处理对草鱼皮胶原蛋白酶解产物的抗氧化活性有显著影响;随着超声时间的增加或超声功率的增大,草鱼皮胶原蛋白酶解产物的抗氧化活性和水解度均呈现出先增加后减小的趋势,且在超声功率100 W和超声时间10 min时均达到最高值,分别比未处理时提高了14.9%和20.6%.In order to improve the additional value of grass carp skin,the collagen antioxidant peptide was prepared by enzymatic method.Alkaline protease as the optimum enzymatic was screened out from 5 proteases.The optimal enzymatic hydrolysis conditions for producing the hydrolysates with the highest scavenging DPPH free radical capability from grass carp skin were determined by Plackett-Burman,steepest ascent path and the Box-Behnken central composite design.The optimal conditions for hydrolyzing collagen from grass carp skin were enzymlysis time 3.0 h,enzymlysis temperature 46.5℃ and enzyme dosage 7025.2 U/g.Under these conditions,the DPPH free radical scavenging rate of the hydrolysate was 78.3%,which was about 100.4% of the theoretical value.Ultrasonic pretreatment has a significant effect on the antioxidant activity of grass carp skin collagen hydrolysate.The antioxidant activity and hydrolysis degree fell after rising with the increase of ultrasonic power and time,it reached the highest activity at ultrasonic power 100 W and ultrasonic time 10 min.Compared with untreated grass carp skin collagen hydrolysate,the antioxidant activity and hydrolysis degree of treated grass carp skin collagen hydrolysate increased by 14.9% and 20.6%,respectively.
