摘 要:采集2017年中国广西壮族自治区部分地区猪组织样品,并对PPV7的流行情况进行调查分析。采集健康猪和患病猪样品的PPV7感染率分别为66.7%和84%。此外健康猪和患病猪PCV2共感染率分别为51.74%和77.33%。二者的高感染率表明PPV7可能与PCV2感染存在一定关系。随机选取3份PPV7阳性样品进行主要结构基因测序,并进行系统进化分析。结果显示,3份样品中全基因组序列同源性为94.1%~99.6%,NS1基因核苷酸序列同源性为96.0%~100%。cap基因核苷酸序列同源性为93.9%~97.6%。与PPV7参考株相比,NS1基因同源性为94.2%~97.6%,cap同源性为87.4%~95.0%。在3株PPV7全长基因组,其cap基因分别编码474,470及469个氨基酸,而作为参考的PPV742株则编码469个氨基酸,cap基因序列变异导致cap编码区域氨基酸序列的增加。Parvoviruses are a group of DNA viruses that are widely infected with various animals.In recent years,a variety of new parvoviruses such as porcine Bocavirus,PPV6and PPV7have been identified by the advancement of molecular biology technology.In This study,samples of pig tissues were collected from parts regions of Guangxi Autonomous Region in China in 2017and investigated the prevalence of PPV7.The PPV7infection rates in healthy and diseased pig samples were 66.7% and 84%,respectively.In addition,the co-infection rates of PCV2in healthy pigs and sick pigs were 51.74% and 77.33%,respectively.The high infection rate of the two viruses indicates that PPV7may have a certain relationship with PCV2infection.The major structural gene and the genetic evolution of three PPV7positive samples were analyzed.They shared 94.1%-99.6% similarity in the whole genome,96.0%-100%similarity in NS1and 93.9%-97.6% similarity in capamong three samples.Comparing with the PPV7reference strain,the similarity was 94.2%-97.6% in NS1and 87.4%-95.0% in cap.By analyzing the amino acids,three PPV7used in this study with the cap gene encodes 474,470and 469amino acids respectively,one or five amino acids were more than the reference PPV742strain.This is the first report that the cap gene sequence variation leads to an increase in the amino acid sequence of the cap coding region.The results of this study provide a reference for the epidemic and evolution analysis of PPV7strains in Guangxi Autonomous Region,China.
