摘  要:新城疫病毒W蛋白是由P基因“RNA编辑”产生的最短非结构蛋白。为了解W蛋白的分子特征,本试验利用生物信息学软件对24株遗传背景清晰的病毒信息进行分析。结果显示,不同毒株W蛋白序列长度各有不同,存在8种不同长度(137aa、147aa、155aa、179aa、183aa、196aa、221aa和227aa),其变异呈现基因型一致性,不同基因型毒株W蛋白长度差异又与强、弱毒特征存在一定相关性。蛋白质分析软件预测结果显示,W蛋白上糖基化位点和磷酸化位点数量与其长度呈正相关性。W蛋白存在稳定的核定位信号,提示其功能活动区可能在细胞核。W蛋白长度的差异是通过改变C端结构域长度来实现的,经二级结构预测发现基因Ⅱ型(中强毒株)和基因Ⅲ、Ⅵ和Ⅶ型强毒株的C端结构域比其他毒株多出1个α-螺旋结构,推测W蛋白可能是NDV强毒株上关键的新型毒力标志。Newcastle disease virus(NDV)W protein is the shortest non-structural protein produced by the“RNA editing”from the P gene.To explore the molecular characteristics of W protein,twenty-four NDV strains with clear genetic background were analyzed through bioinformatics software.The results showed that the length of W protein differed among NDV strains with 8different lengths(137aa,147aa,155aa,179aa,183aa,196aa,221aa and 227aa).The variation showed genotype consistency,and the difference of W protein length between different genotypes was correlated with strong or weak virulence characteristics.The protein analysis software predicted that the number of glycosylation sites and phosphorylation sites on the W protein was positively correlated with its length.The W protein has a stable nuclear localization signal,suggesting that its functional activity region may be in the nucleus.The difference of W protein length is achieved by changing the length of C-terminal domain.The prediction of secondary structure shows that the C-terminal domain of genotypeⅡ(moderately virulent strain)and genotypeⅢ,Ⅵ and Ⅶ virulent strains has more one alpha-helix structure than other viruses.In summary,these results suggest that W protein may be a key new virulence marker on NDV virulent strains.
