摘  要:为查明吉林省长春市张二路鹿场致鹿死亡病原菌,对分离出的1株病原菌进行形态学观察、生化鉴定和16SrRNA基因扩增,并用NCBI blast及MEGA5.05进行序列分析。结果显示,从剖检死亡鹿肺脏中分离出的病原菌为大肠杆菌。采用玻片凝集法鉴定其血清型为O102型大肠杆菌,命名为Ed-102。在此基础上,为确定分离株的生物学特性,对分离株完成运动性分析、生物被膜检测、人工感染小鼠试验及药敏试验,结果显示分离株Ed-102可以在LB半固体培养基上形成运动圈,生物被膜形成能力较弱。人工感染小鼠试验显示小鼠感染12h后开始死亡,剖检小鼠肝脏、肺脏及脾脏有不同程度出血,其对小鼠的LD50为1.20×107 CFU/只。药敏试验结果显示分离株Ed-102对氨苄西林、哌拉西林、氨苄西林/舒巴坦、头孢唑林、头孢吡肟、头孢哌酮、头孢噻肟、头孢曲松、头孢他啶、氨曲南、卡那霉素、链霉素、四环素、西诺沙星、萘啶酸及氯霉素高度耐药,仅对于阿米卡星及亚胺培南较为敏感。In order to determine the pathogen causing deer death in Zhang Er-lu deer farm in Changchun,the isolate was subjected to morphological observation,biochemical test and 16SrRNA identification.And the amplified sequence was analyzed with NCBI blast and MEGA5.05.The results showed that the pathogenic bacteria isolated from the lungs of dead deer was Escherichia coli.The serotype of O102 Escherichia coli was identified by slide agglutination method and named Ed-102.Basis on this,kinematics analysis and biofilm detection,drug susceptibility test and artificial infection were completed to determine the biological characteristics of the isolate.Test of artificial infection showed that the infected mice died after 12hours and the lung,liver,spleen and kidney showed hemorrhages in different degree.The LD50 was 1.20×10^7 CFU/mice.The drug sensitivity test showed that the isolate was highly resistant to ampicillin,piperacillin,ampicillin/sulbactam,cefazolin,cefepime,cefoperazone,cefotaxime,ceftriaxone,ceftazidime,aztreonam,kanamycin,streptomycin,tetracycline,cinofloxacin,nalidine and chloramphenicol,but sensitiv to amikacin and imipenem.The purpose of this paper is to identify the pathogenic bacteria causing deer death and provide basis for clinical treatment.
