摘  要:将150只雏鸡分成5个组,每组30只,1、2组分别为阴性对照组和阳性对照组,第3、4、5组在7,14,21日龄时分别肌肉注射重组AMA-1蛋白50,100,150μg。在28日龄时,除阴性对照组外,每只鸡经口灌服8×10^4个E.tenella卵囊,在试验的14,21,28,35d时每组采集5只鸡的心脏血液收集血清。结果显示,第3、4、5组各期的血清AMA-1抗体水平均显著高于1、2组(P<0.05),尤以3、4组提高幅度最大,与5组比较达显著水平(P<0.05)。血清IgG、IgM、IgA含量也呈一定升高趋势,尤其在试验35d(即攻虫后7d)时,第3组血清IgG、IgM、IgA含量显著高于第1、4组(P<0.05),而血清IgG、IgA含量也显著高于第2组(P<0.05)。免疫各组血清T-SOD、T-AOC、GSH-Px活性较1、2组呈升高趋势,其中在试验35d时,第3组与第1、2组比较差异显著(P<0.05),而T-SOD、GSH-Px活性也显著高于第4组(P<0.05)。结果表明,适量的重组AMA-1蛋白能够刺激机体产生特异性抗体和免疫球蛋白,提高机体的体液免疫水平,增强清除机体氧自由基的能力,从而起到抗球虫的效果。In order to determine the effect of AMA-1recombinant yeast protein fromE.tenella Hebei strain on the humoral immunity and antioxidant capacity of chickens.150chickens were divided into 5groups,with 30chickens in each group.Group 1and 2were negative control group and positive control group,and group 3,4and 5were intramuscularly injected with 50,100and 150μg recombinant AMA-1protein at the age of 7,14and 21days,respectively.Except for the negative control group,each chicken was orally administered with 8×10^4 E.tenella oocysts at 28days of age.Five cardiac blood samples were collected from each group to separate serum at 14,21,28and 35 days of the experiment.The results showed that serum AMA-1antibody levels in groups 3,4and 5 were significantly higher than those in groups 1and 2(P<0.05),especially in group 3and 4,the increase was the greatest,which was significantly higher than that in group 5(P<0.05).The contents of serum IgG,IgM and IgA also showed a certain increase trend,especially at the 35th day(7 days after the attack),the serum IgG,IgM and IgA levels in group 3were significantly higher than those in group 1and 4(P<0.05),the serum IgG and IgA levels were also significantly higher than those in group 2(P<0.05).Serum T-SOD,T-AOC and GSH-Px activities in immunized groups were higher than those in groups 1and 2,and at the 35th day of the experiment,there was a significant difference between group 3and group 1and 2(P<0.05),the activities of T-SOD and GSHPx were also significantly higher than those of group 4(P<0.05).These results indicate that an appropriate amount of recombinant AMA-1protein can stimulate the body to produce specific antibodies and immunoglobulins,and improve the body's humoral immunity,and enhance the ability to scavenge oxygen free radicals,so it has the effect of anti-coccidia(playing a coccidiosis-resistant effect).
